Just wanted to give an update on my experience with this tek. Super easy to whip up. I checked them this morning and there is no liquid on the top layer of agar. I have put them in the fridge with the tinfoil still on to firm them up some more. I read somewhere that you shouldn't use them til 2 days has passed??

It doesnt matter, people just leave them a couple days in their SAB usually to reduce the condensation if there is some before inoculating.

This is mainly to see if any poured plates were contaminated during the pouring process. Bacteria would show up in that time, and mold likely would in two or three days as well. even with no pours i wait a couple days to let the condensation evaporate or reabsorb so i can see into the dish better once the mycelium starts growing out. I've never actually waited with my pour dishes (but i may now) as they have less condensation and i've always been paranoid about leaving unwrapped petri dishes for too long, though recently i even read RR saying he just leaves the sleeve on after cooling and folds and tucks it underneath and that's keeps them clean until he needs them. SO, my last round of transfers did not require all the dishes i poured so i did as rr said and my paranoia was settled some. They've been there for close to a week now so i can take a look for any bacteria or mold before i use them. ALSO I found some plates i forgot about that had been poured and left on a shelf with no covering and unwrapped and they all looked just as they did two+ weeks prior when they cooled. I still threw them out because they were exposed to open air for so long and any dust/spores that had settled on them would contam them as soon as i openned the dish. STILL Judging from those dishes i can see that my pouring process is ok AND i shouldn't be so paranoid about not using the poured petri dishes immediately(as long as they are wrapped or in a sleeve). Anyways main point is if using no pour i like to wait a couple days for the condensation to reduce. basically Quote:

so another question i have is when storing no pour plates for future use is it best to take the tinfoil AND PAPERTOWEL off once they've cooled to make sure the filter doesn't stay wet? (I'm using SFDs as my filters)

Nope I leave it on, the paper towel will easily absorb any moisture and keep it away from the filter. I have left them wrapped for a week and they are bone dry when I unwrap them again. I also never take off the paper towel. I use it as my lable and write any notes on it.

I was worried because when i take them off once they've cooled (the next day) the paper towel is still damp. I'll try keeping it on next time and see if they are dry after a week. do you think i can stack pasty plates (or ziploc rounds) while colonising or will this possibly cover the ge too much or cause wet filters from the one being stacked on top of the filter(sfd)?

No I use poly covered in micropore tape but It shouldn't mater. You will still get enough GE.

if I make up a bunch of these using 1/4 pint jars, how long could I store them before use? and would I put them in the fridge, or just keep wrapped in foil and placed into a empty tote? I'm wanting to try my hand at getting a monoculture.

tom if your using sfd's, try it without foil and PT, i know i dont need to cover my grain jars when i pc with sfd's so..

I tried this before and they all had more condensation than the ones with PT and TF and they also had puddles of water on the lid when i took them out of the pc which may have partially absorbed through the filter into the dish(may be because of the shape of the ziplocs). I didn't use those ones because the condensation never reabsorbed and the dishes (almost) all had a puddle on top of the agar.

I usually make a few extra and just leave them in the corner of my sab until I am ready to use them. I think the longest I have left un inoculated plates at room temp was about 3 weeks but I'm sure they would last longer.

Ive found a lot on info on making agar and how to avoid contamination but not much info on what to do once I have growth. Anyone have a detailed log or tek?
