Yes, you can take a very thin piece of clean tissue from inside the steam and place on agar in a still air box.

I did an experiment to see is open air cloning was a disaster. All I was attempting was to get the tiniest bit of Myc growth to cleanly transfer asap. Result wa I beat all the contaminants and grabbed my culture.

Hey, new poster here.
How do you prevent condensation from building up underneath the lid? I drilled the hole on the side but I still ended up with a fair amount of water built up inside some of the mini-rounds when I took them out the PC.

Normally all no pour should be done container upside down 🙃 except when dumping excess water. Carefully dump any excess water in a sab. Excess water evaporates in 5 days if you do nothing.

Do we need to cook in the pressure cooker at a certain pressure? Or does it not matter? Just using the pc as a sealed steaming unit to sterilise the contents?

15-18 psi it will be sterilized at lower psi it just takes longer

Thank you very much, the reason why I ask is my pressure cooker will only reach 8 psi, so I'll just add another 30-60 mins on the time.

There's a difference in a pressure cooker and a pressure canner. Pressure cookers do not get hot enough to sterilize properly, but a pressure canner can and has been used successfully as a sterilizer and will reach the temperatures necessary to sterilize grains, surgical instruments and much more.

I'm just wondering about this in general, how many plates do you guys have before beginning? I always seem to find myself in need of more when I need to do transfers. And how many transfers should I calculate in the whole process?

Some agar I have made has a thin layer of liquid on the top, I have a feeling that something happened when I was cooling them down and some humidity got in there and made some water. Does this mean this batch is ruined? Or can I pour this excess liquid off of the top and itll still be ok?
I put some droplets of spore syringe in there before I realised that there was excess liquid, will that extra liquid cause it to not grow anything? It's been about 3 days and I can't see anything at all in there.
I think next time I might use something to scrape the spores off the foil with instead of using a spore syringe. Would a qtip suffice? or i've also made a very dodgy inoculation loop. That might be better.

when you go to inoculate the dish, turn the plate over first to let the moisture drip off. what remains should evaporate through the tape. i store my damp plates upside down.
spores can take a while to germinate. not sure if the water will prevent this, but i reckon it will be ok. again, consider storing upside down.
bodhisatta made a sweet how to make a noc loop thread. find it. swabs work, if you sterilize them first. wrapped in foil, in the PC for a little while.
