Just wanted to say on making an awesome agar tek for a noob to start off with!! I made 7 pasty plates the other day and did a transfer from my first spore print the next day. I see some growth going on there... I saved 3 of em for some tissue samples. After doing the tissue samples, I realized that 7 ain't gonna do it (I poked a hole in one making the ge holes). I need MORE mini's!!!
Once I get my isolate, after however many transfers, I can make a master to do g2g, right? How big of a wedge would I need for a master quart sized jar? If it makes a difference, I'll be using wbs. Thanks and great job Pasty!!!

You don't have to wait for an isolate, clean strong growth will often get the job done if is ms too. So usually I do about 3 transfers then go to grain. A wedge the size of a quarter will work but larger ones or a few small ones will get the job done faster.
For making a fast master, the tiger drop is pretty easy Its a lot of inoculate so even typically slower species will get done quick

look at the recycling code on the bottom if it is 5 or has the letters PP it will work. Glad or ziploc containers are all good.

They will not melt if you make sure there's water in the cooker! (Recycling code 5 plastics)
If you run out of water... prepare for melted plastic!

Sure, Violet uses them all the time. I have used them but I find they take up too much space in both my PC and fridge. But go ahead and use em if you prefer or its all you can find.

They do take up more fridge space (I only need a few in the fridge tho) and fewer do fit in the PC. But I can use them for literally Every facet of my grow, from culturing to the full grow and more.

After sterilized, simply remove the containers from the cooker into the sterile workspace, and in a single fluid action with both hands, place one hand's thumb and finger on the 2nd container from the bottom:
 ... lift, leaving the bottom container as you set the others next to it:
 ... while taking your other hand to the top lid:
  ... and placing that top lid on the lidless container. Repeat until placing down the top container which is already lidded. It's easy and quick, much moreso than lifting stacks of single-use plastic petri plates and pouring warm/hot agar into them in turn.

Try less agar in them and a slightly stiffer mix. With food grade agar being somewhat inconsistent in gel strength it will sometimes require a little tweaking to get just right. Also be sure to let the PC completely cool and then some before removing them. If the rounds are even a little warm inside before you open they will get a lot of condensation.

Yeah try a few things until you get it dialed in. I have also found that even the recipe will change gel strength. MEA ends up firmer than PDA and cornstarch is firmer than MEA. Just need to experiment a bit till you find what works for you. Also try having less in each round. Its dry where I live so I might be able to get away with a little more than someone who lives im Florida for example.

I had plenty of water in my pc and still got some shriveled containers. The glad ones pasty suggussted. I might try very small mason jars nest time, only drawback being the lid isn't so easy to remove.
