Dude 30 hours is not a long period of time at all by the way, people in the RustyWhyte thread were stoked that someone got germination of spores on plates in 2 days (so 48 hours). It can take a week, longer if the spores are old/dehydrated.

Yeah 48 hours is almost unheard of. Like record breaking. If I had germination in 30 hours I would suspect it was mold.

Followed the instructions but after pressure cooking had a few glad containers get crushed and lids are loose now. They have holes donβt know why some got crushed. I did snap down the lids for pressure cooking. Was I supposed to leave the lids on loose when pressure cooking? And should I wrap the loose lids?

Iβve never had any crush before. How many did you put in at once time and how long did you PC them?

Upload a pic of your pasty plate. Either the hole is too small or clogged. Or user error in dropping pressure by removing the weight.

I made a labeling mistake! Argh!
I have two ziplocs of pasty plates, one has been PC'd one has not, in the fridge. I didn't label because I had 7 pc'd and 6 not, easy to tell apart. Then I had a random contam and had to transfer, grabbed a PC'd plate and transfered.
Now I need/want to do some transfers and I do not know which is which. My question is, is there anything wrong with PCing a pasty plate twice to ensure all of my plates have been PC'd?
Also, how long do you leave a plate and wait for MS inoculation? I noc'd 16 plates with treasure coast spores from a vendor July 15. 8 of them have already been transferred, 8 of them show nothing.

yes you can PC them twice no problem man!
could take 4-5 days to see growth and up to a couple weeks. if sooner than 3 days maybe contams so watch close.

Awesome, thanks for the quick reply, pressurizing the cooker now.
Yeah, these have been going for 11 days, I guess I will leave them until 30, unless I need the plates for viable, growing myc. I must have been really lucky with my GT on PDA, I had strong enough growth to transfer in 5 days! MS noc'd 7/15, transferred 7/20, and how one of the second transfers is looking today. Ps these containers suck, I am sorry.

only that doing it twice wastes time and energy. But so does growing mold so I would do it.

Until they germinate or the other ones need a transfer. There have been a few times that inoculate 8 plates, 3 germinate then they need transfers but im too lazy to cook more agar so i just transfer to the plates inoculated on the same day

hey guys, day 5 of my pastys here, how are they looking? are the black specs on the underside harmless? search bar says they might just be spores but i dunno... ill be making grain water agar when im ready to transfer my isolates once i have enough growth for ~10 clean clones. the condensation makes it hard to get a good pic, sorry bout that

sorry i should have clarified, i used malt extract not tater flakes! ill be using grain water for my 2nd round isolates.
