First time trying to inoculate was a goddamn disaster lol. I think my plates are too wet. After sitting in the pressure cooker for several hours to cool, 2 of them still had the consistency of water, while the other ten were solid and could be held upside down. I knocked up 3 of the dryer ones as a test, and left all the others to sit. First time using my SAB with agar though and I'm pretty sure my spores just ran all over the place with the puddled water lol. Definitely think I'm going to leave my jars out for a few days to dry before I mess with them in the future. I'll check back in a couple days I guess. See if I have any growth and if my other plates lose some condensation. Do you think I should wrap all the un-inoculated plates in cling wrap if they are only going to be in there two days? Trying to manage a roll of that shit in a SAB with gloves on and everything soaked in alcohol is frustrating as all hell. This is with ball jars so they are sealed tight as is.

Water on the surface is a good thing, it helps spores germinate. I'm not sure why 2 of them didn't set..

I'm guessing I didn't get the premix MEA mixed well enough with water before I poured the plates, and those two had too much water relative to agar. I'll probably bump up the MEA to water ratio a bit in the next batch and mix it a while longer.

Was the agar completely cool when you inoculated? Sounds like you did it right out of the cooker.

Everything was cool to the touch. Waited about 5 hours after killing the heat. Certainly on the early side, but there's definitely something fucked up with those two that haven't set.

No, unmodified. Maybe I backed the lids off too far on those two.

I doubt it. Your first thought sounds the most plausible.

That doesn't sound right, agar is ready to pour when it's uncomfortable to touch but not scalding you, in 15-20min minutes the poured plate should be solidified.

I did this tek today but instead of agar I used brown rice flour substitute from another tek. I'm excited to see how they turn out! Will post results when I see growth. Only bad part is it may be a little harder to see being that the BRF is white.

beerface said: I followed these directions for my first Agar usage. I bought the glad mini rounds as stated. I filled them with my Agar mixture about the same as the pitcures. THEN SNAPPED THE CAPS ON, with the micro pore tap. wrapped them in tin foil. Then I PC'd them for 40 mins at 15 PSI.I also didnt bring them up to temp blazing fast, it took about 20 or more mins to get to a boil, then build pressure. I waited about 5 hours and everything was pretty cool. nothing was close to being hot.
So when I put them in my SAB and unwrapped these bad boys I noticed the caps didnt snap on or off any longer, Its like they were just suggly on top, they didnt seal any more, like they changed shape. what happened? did I use to much PSI when I pressure cooked them? im very disappointed as I wanted to start my process today. I still took about 7 of them and put a drop from my syringe on them just to see what happens. But i know they arnt sealed correctly and need to know what I did wrong, so I can make new ones.
so its been a week now since I have done this. There is no sign of growth or contamination on any of my containers. how long does it take for any growth to happen? I also used/prepared this agar mix and added a couple drops of red food coloring in it

The lids will do that. You just need to handle them very carefully.
Germination may take a while. What are your temps like where you're keeping them? Cooler temps will make things take longer in general

is there a reason you dont like this brand? and does any one know a better premixed brand? I find it easier to get premixed as stated by rogerrabit in his videos too.

I have never used premix, but I hate the idea of not being able to alter ratios
