Hey man 20 mins at 9psi is more than enough to sterilize those plates next time
I don't agree. I have seen bacteria running short times with lower psi. Elevation and type of media also plays a role. Lab grade media usually has smaller populations and so can get away with less time.

This is most certainly true, I live at around 2500 ft elevation, so water does not boil at 212 here. It's lower. So with that in mind I try to run at least 17 psi to get to normal pic temps for the guys running 15 at sea level where the atmospheric pressure is greater, something for all to keep in mind, be cognizant of your altitude and its effects on boiling points

I wish I could change my psi :/ Which is why I triple it. I like these new holes in the lids though. Micro held up good.

Hmm interesting. I must have just been getting lucky all this time then My electric pc that I use for agar is 11 psi and I only ever pc for 20 mins. I assumed I should treat PDA like any other liquid media, no?

higher populations, yet pasty is well above sea level. 20 minutes is fine at a low enough altitude

Yeah I should really be more aware of that damn altitude factor when giving advice

im super newb to agar and trying my first attempt thanks to this tek. dont laugh at me here but do i use my spore syringe and just let a couple drops through the hole in the top after it has cooled overnight? if so, would i stab through the tape or peel it back to put the LC in?

no dont stab through the hole. you have to remove the lid to inoculate it.

Just to be clear, are you trying to put a spore solution onto the the agar, or a liquid culture? I think your talking about a syringe of spore water correct?

yes, for ten years i have always used a syringe into a jar of PCed WBS. This is my first time doing anything different so im pretty clueless here.
Something is telling me a spore syringe (water and spores) is different than a liquid culture...?

Yes they are different things. I figured you were talking about a spore syringe though. I haven't messed around with lc's yet. But Munchauzen answered your question. Just be sure to put a small drop on each plate. First time I tried I oversaturated it, "thinking it would be better" and fucked it up. Good luck with everything and welcome!!

Thanks much G! Ive literally been reading about agar for a month, finally found the powder in some oriental market downtown, and now that im ready to try i thought there was an entire step i was missing about getting the spore to the agar.
I suck at terminology here, i feel like a caveman when i read. Time to find a shroomery dictionary

Your welcome, Same thing here, oriental market downtown haha. It took me a week to figure out what everyone meant when they said pasty plates. I thought it was some kind of Petri dish that had sticky paste Would have been much easier if I just asked haha Dont be shy when it comes to asking questions.

So petri dishes can't be used for a no pour tek?? I am slowly making my way through some of these type threads and I prefer the easy as fuck ways, not necessarily the EAF teks, but am always looking for easy, lazy, effortless ways. I have disabilities that present definite challenges to what most find easy, if not tedious and laborious.
I was gonna try and use this style and use glass petris. What is the problem? boiling over? Is there any chance of bringing the pressure up very slowly and preventing this? Or is this even the (only?) issue?
Hope this thread ain't totally dead. And yes I do know how to UTFSE, but it don't always find what you're looking for.

correct. boiling over. they are too shallow. bringing up the pressure slowly would make no difference. its still going to boil and its still going to spill out. don't do it.

I have heard of some users finding tall ones that they got to work but it's not at all worth the hassle
