Jonhova said: I have had 3 different batches of agar go bacterial at day 4 after pcing them. Really frustrating me to no end. Whats strange is i had done about 5 rounds before this and had great results.
I'm not an expert with my pc by any means but i was getting perfect agar using an inch of water, letting it steam for 10 minutes at low/medium heat and then adding the 15 lb wight for 30 minutes. I used the recipe here to a tea(spoon).
I tried again last night, letting it heat up very slowly (25 minutes.) Then raising the heat to medium high. I let it steam for 10 minutes, and then added the 15 lb weight. I reduced the heat to medium for thr 45 minutes.
This last batch's paper towels are half damp and a handful have wet micro pore tape. They all have aluminum foil coverings. I had them raised on 2 ceramic plates and the trellis. 3 quarts of water.
I had them about 3.5 inches off the bottom of the trellis.
Can anybody tell me how to bring the heat up to avoid splashing yet hot enough to sterilize the agar and grain jars?
make sure your holes and micropore aren't breeched

@tryptkaloids:
The paper towel and the double filters on some were intact and slightly damp. Apparently that doesn't matter as much as I thought it would by my results. no signs of contams on the last batch.
All the teks I read pointed to a rocking motion on the weight to signal 15 psi. Even at high heat my weight didn't budge at all on my 16 quart presto (per bods tek I taped quarters on the top making it even harder to identify movement. STUPID ME).
The heat on my pc continued to rise spurting water the whole time, until the blow out valve extended all the way out. At which point I became confused, lowering the heat.
......It finally dawned on me that the PC with the weight on should not be spurting water and that by continually venting as I had been previously I was doing it all fucking wrong and running the risk of going dry.
How to pc a rocker presto 16 the right way: add 3 quarts of water, raise the pasty plates up out of the water, bring temps up to vent, leave it venting for 10 minutes, place #15 weight on, when you see steam venting again reduce heat slowly until no steam is escaping.
Kinda weird everyone's write-ups go by the rocker moving, when steam would be much easier visual cue imo.

If iv found myself with too many cultures to work with at once, can i store pasty plates in the fridge for up to a couple months to slow down the growth on the plates? Or am i gonna have to store them on slants or something?

Yay! I'm glad I could help. Someone was really nice and helped make sure it went well for me the first time and I'm glad I could pass down the information.

you want the water near boiling when you put the lid on. the steam is to equalize pressure and insure an even cook, the higher the pressure in your pressure cooker when you put the weight on the sooner you can turn the heat down. with the times illustrated in this tek venting isn't 100% necessary but it is still recommended. your problem was too much pressure too fast causing the water to expand rapidly. don't worry too much about raising temp too fast just let pressure build before adding the weight and starting your timer. with 17 psi from faucet temp water I'm not surprised you couldn't get to pressure

TFW widemouth jar rings make your PC's rack uneven and you don't want lopsided plates, but you're too lazy to grab four reg mouth rings so you just load the PC with its thin-ass rack and hope there's enough water.

Holy shit man. get yourself a couple of trivets.
I'm so glad mine came with two trivets. Also I'm glad I have a pressure gauge to monitor it and it's almost dummy proof. So far the only bad thing that's happened is I left the steam valve open last night and had to leave my pot on longer which caused my hot plate to die.
Also tell me that's not melted glass cause glass melts at 2800 f. It's plastic right. I don't need to have any nightmares about my PC being a crazy hot inferno that can melt glass.

Me too! To prop the tray above the water a little more. I made some Pasty BRF, noced them with MSyringe. I grew these alongside spore streaked petris. The substrates germinated much better exploding into growth, I just isolated a rhizo from one of them and put that on agar. I am still isolating tiny confused masses of genetics waiting for them to show some consolidated personality from the petris.. I think that may be my thing, Spore injected pasty substrates plates to start a spore based isolate then put that on agar, Of course only if you are too rushed or uncertain about print quality to just wait for pins.
BTW The BRF/RFplates and cakes are performing the best so far.

No glass, it was thankfully just Glad rounds. I'm sure glass would have been a real PITA to clean. I did another batch, with proper elevation and water, and discovered that the overpressure seal on the lid is warped. That definitely played a factor in my Pasty Plate disaster, though I still think the water level would have been too low regardless.

Veggie steamer is not a bad idea...I'll have to test that. I do plates in a lil 6qt tho, so it might get kinda cramped. I have a 22qt, but usually save it for spawn/subs cause it takes longer to heat up.

the glass wouldn't melt it would just shatter, it wouldn't be nearly as much of a PITA as pasty plates

Good point, but I gotta say the P. Plates cleaned up pretty easily. Smoke detector started going off before they got too bad. There is a lil crustiness left behind, but nothing worth scraping off.

I didn't have a smoke detector when I did it, I threw the trivet away instead of dealing with that shit

Could someone please help walk me through the process of pressure cooking the plates?
I used the search bar and looked through the comments on this post and wasn't able to find much.
Last time I tried this tek I burnt the bottom of my pc and the tin foil that I wrapped my plates in. Any help would be greatly appreciated!

Thank you. I will do this! I have one last question:
Quote:
Now you just gotta PC them for 40-45 mins
Should the whole process take 40-45 minutes or should I keep it at 15 psi for 40-45 minutes?
