i just used the water amount called for useing the grain soak water and the agar.

Sometimes it's really more of an art than a science. I weigh my agar and approximate the rest. Pretty hard to go wrong with PDA/GWA.

For a lc or spore germination I use just grain water and agar but for rhyzos I use 50/50 or 25/75 gw/water

A couple days ago i made an attempt at pastys agar tek which seemed simple enough. I did have a few problems i wasnt sure the best way to get past....
First issue was that the natural agar i used took entirely too long to dissolve. I think i lost alot of water in the process which made the agar overly rigid. I put the stove on low as instructed but i had alot of particulate until i turned the heat up 30 minutes later. Cooking it for nearly an hour. Seemed wrong to me. Should i start with higher heat or perhaps use a lid?
I did bring the cooker up slow to a boil then put the weight on the cooker lid and ran the pc for 40 minutes at 15psi. I turned the burner off then which may have caused the condensation in the container. Perhaps i should have turned the burner down then off. It seems some of the moistute has let over the past day.
The last issue i had was getting to a decent tissue sample. Half of my stipe i had wiped with h2o2 and half i didnt to see if it made a difference. The core of my stipes was alittle drier than the outer layer. I flamed my scalpel and let it cool before removing the sample. (Is there a better way than just waiting? I tried dousing the blade in the agar before cutting the sample...not sure if that's ill advised.) After very carefully scraping my sample from the center of the stipe, i popped the lid and tried to stick it to the overly rigid agar. I checked them today and most of the samples have shriveled and bruised. Is this normal? In pics i see nice little white pieces of myc on the agar surface. Any recommendations? I really need these clones to work or im starting from ms again

Is the sample suppose to stick into the media? Or sit on top. My media was like a very still jello

the point is to have a 2-D surface for your sample to grow out on so you can cut out the good growth and transfer to a clean plate. you don't want your mycelium to grow into the agar, it should be solid so it will grow on top because that's why we even put agar in

Unfortunately theh still look the same. How long does it generally take to see a change on clone plates?

how long has it been? it generally takes 3-5 days to see recovery from cloning depending on the culture's genes and cleanliness

Only 3 days so who knows. I making 12 more now and have purchased a coffee grinder and scale to better my result. I will try clones in half with samples i had to refrigerate and the other half ill try with a print. Will post results at end of week

This is my first time fucking around with agar. I took 12 clones from all over my fatest Golden Teacher fruit to see which would grow the fastest. Heres's a few pics of 3 good followed by 3 bad clones taken on 5/28.
Only 1 of the 12 has shown green mold. The rest of the weaker clones seems to be either not growing at all or a weird faint wispy white growth covering most of the agar.
I love this fucking tek for being simple and cheap.

No. Use less grain water or brf or whatever you mix into your agar.

Using less agar will effect the consistency, if you want less nutrition use less potato flakes and still the drop of Karo or honey
