@maxpassin Also, don't add natural honey to the agar mix, it has antifungal properties and it's going to slow down your cultures a lot. If you can't find karo or the fake honey that is good for the tek, it would be better to not add any sugars, only potato flakes. You can also go to a health food store and buy sugar in the raw (pure cane sugar) but keep it under a 1℅ ratio.

@Josex Definitely didn't know that. I thought the purer the honey, the better. I do have some raw cane sugar (it's almost like a solid brick) laying around, I'll probably that.

Yep, it solidifies after a time, but it crumbles up easily and it's still good to use.

Hahaha, I made the same mistake with my first 10 petris. Except I didn't fill them with agar. They came sterile and my dumb ass wanted to sterilise them again.

IDK what im doing right but I use glass petris and never have a problem with them boiling over in my pc.

Maybe if you stack them, there is enough pressure on the lids that they don't come loose? I'd still rather pour petris.

I can vouch for glass petri's not boiling over as well. Must have been some really shallow petri's that perpetuated that myth that has taken root as fact.
But I also prefer pouring, after sterilizing glass plates in the oven @ 350 for an hour. If I pour at proper temps, the glass has little to no condensation, allowing greater visibility.
But glass plates are expensive and not available at the corner grocery store like these pp5's that Pasty uses. Pasty's Agar Tek is a great system for getting the job done with a small investment.

I agree. Plus it provides a nice microclimate for growing invitro off the agar if you want a quick clone.

I used honey in my agar for a long time with great results. I did some research on the antimicrobial properties of honey. Studies have shown that bringing honey up to High temperatures kills almost all of its antimicrobial properties. So after the PC cycle it's basically just sugar.

I was having problems some time ago with my cultures being very slow and I can't remember the exact conversation here in this thread but I concluded that I added too much natural honey, then I tried adding a little less but the cultures started from spores were still slow. That was my experience but I can't be 100% sure to this day that it was the honey, all I can say that SITR seemed to work better for me with the same nute ratios.

Your problem could have just been that you were using too much sugar in general. Mycelium grows slow as fuck when there is too much nutrition available.

That's very likely. The good thing about SITR is that you can weigh it out but the natural honey I had didn't have a drip dispenser (or whatever the name in English) and one day I weighed out a single drop of natural honey from a spoon and man it's fucking heavy, no wonder I fucked up the recipe. Then I did a search and read RR saying that natural honey has indeed antifungal properties, so I stopped using it.
But what you said about honey breaking down into sugars after the pc cycle makes a lot of sense, it would neutralize its antifungal properties.

Yeah raw honey is antimicrobial but things kinda stop being raw when you PC them. I use dextrose jn my afsr these days but I still make honey LC once in a while.

How many PC runs is everyone able to get from their plates?? I've been able to do like 6 but feel like the jars aren't closing as tight.. I may just toss em and get new ones.. For the price why not

100s. I've still got the first ones I picked up almost 3 years ago and they have been run every week or two since then. Yeah the lids get loose but it's better that way. They are much easier to work with once they are broken in. You just have to be mindful of the looser lids when you handle them.

I've had at least 8 with no sign of wear yet They're in the PC right now.
I also started saving and using my marinara sauce containers from Little Caesars. They're great. They are the same height but have a bigger diameter which makes transfering easier
