Quick, any downside to prepping my agar in the microwave in a plastic measuring cup instead of in a pot? Honestly it's because the pot needs to soak a bit and I wanna get this done now and I boil water in this measuring cup all the damn time. Would be easier to pour into the plates that way than from a pot.
So, other than maybe some vigorous bubbling for a minute, no real reason not to right? Still plan on PCing normally after that.

When you said you go on the wetside for MS (did you mean for MS solution or print?) germination, would you say for a regular batch that is 25g mea to 500ml water to do about 24g mea to 500 ml water instead? I know you use pda but maybe you can relate?

Definitely. Enough water to complete the run without boiling dry, this can damage your PC. If the PC came with racks use them or you can make some out of cheap baking or pizza pans cut down with holes drilled in them with canning rings or foil balls under the racks so the plates sit level and out of the water

Question...could you PC pasty plates for 90 minutes along with grain jars or is that too long for them?

No. That long would likely cause excessive caramelization. The only thing I might try that with would be grain soak water but even that might be too long for it.

How about 60 min?
And if I left my pastyplates in the pc for over 24 hrs after sterilizing should I pc them again?

60 is fine for grain water but a bit over kill for anything else, if you use the recope in the OP it will be fine tho. Your plates are fine after 24 hours.

So will the plates that were pc'd for 60 min not preform as well? I did that just as precaution and for altitude. I'll stick to the recipe in the future

Use whatever recipe you want. Changing recipes is a good thing. But if it's got a lot of sugar then carmelization is a risk. So don't over PC. I PC grain water for 45-50 min. Malt needs less.

I doubt 15 minutes more is going to make much of difference. If anything its just going to further guarantee sterility.
Definitely listen to pastey over me though hah.

Just thought I should say thanks for this technique. It is what made me comfortable with working with agar. Now I've made masters and even use real Petri dishes all prepared following these steps

Just got 9 wedges in the mail (all edible) and immediately put them to PastyPlates. I feel like a real mycologists working with agar and it is truly due to you (and a couple other culters)- my understanding is deeper, and my passion is more fully bloomed thanks to this technique. And while 9 months ago sterile work scared me- now it's my best friend and I can't wait until I do more.

It's truly because of this thread that my passion is so intense- why all of my bags right now have been cloned from store bought blue oysters following this tek- and I have several other cultures due to the Gormet thread that I almost (almost) don't know what to do with them all, plus some local badasses hooked me up with some aloha cultures (which if you want a wedge pm me and we will trade) - all in all sterile agar work is my favorite part of the week.
Can't thank you enough Pasty- you made it approachable, affordable, and easy.
