Those are my favorite. I can't get them anymore but luckily I have abut 100 of them and they hold up really well threw many many runs.

^^ @ CapMeh Oh, that's bacteria for sure dude. The water in the plates is from condensation, so there won't be any nutrients to speak of in it.
Often times some of my clone tissue samples get surrounded in liquid that isn't as obviously bacteria as what you have, and never get rolling on the plate. I just take multiple samples, they seem to have about a 50% success rate. Supposedly there can be bacteria in the stipe tissue.

I spawned some popcorn casings and had double-overlay cause I didn't put enough hydrogen peroxide in my automated aquarium mister. I only got one mushroom so I cut off the head part where the seeds fall from and put it in a jar of LC and sprayed it all over a tin of PF cakes I made with gravel, cardboard, and bisquick in my microwave. I think it will be good cause B+ is so potent. Triggered yet? Only a square would say "a cube is a cube."
No, this does not look right...

This third plate looks a little different than the others; My first thought was bacteria (maybe my knife was dirty for this plate or something), but then I thought maybe it was just water that had gathered around the tissue, water that might have had a little malt suspended in it and the myc is colonizing it like a mini LC? I can't really see too well but that looks like bacteria to me. Normally my clones run much faster than that. There are variables to consider though. How much nutrition is in your agar mix? What temperature are your plates colonising at? Genetics is a big one too. Some cultures are a lot faster than others. Also if you clone from a mature fruit it will move slower than a clone from a pin.

It's because spores are not magic bullets, they fall straight down usually. They definitely won't blow in sideways then up then over then inside.

The ziploc 8oz ones? It would be grate if they'd be 4oz. i was thinking in using SFD instead of micropore, don't trust in micropore.

I've started using SFD on some of mine but honestly micropore works fine I don't even replace it from run to run.

I guess ill try micropore. How many layers you use? And you use that papertowel Pasty uses in his Tek?

I use two layers of tape with no paper towel and foil on top. I used to use paper towels. Then I didn't one run and haven't looked back. I think I've only ever had one pasty plate contam prior to opening it in my SAB and I'm pretty sure it was something else.

That's how I roll too.
I got my first pre inoculation contam in a pasty plate a few weeks ago but I remember the lid popping open while I was taking off the foil. I actually transfered to it thinking I had used green food coloring in that batch but on closer inspection the entire plate was green with trich spores.

Thanks for the very detailed play by play. You are making it easy for me to try, and it is appreciated.
About contams standing out with colors. I saw a petri dish pic with clear gel. I thought I could see all the bad stuff cause gunk wasn't clear. But you say actually things could hide, and i believe you. So really the darker the better? What about Black instead of green food coloring?

The mycelium doesn't really like black food coloring, starts absorbing it and it changes the color of the mycelium. Black is too high a concentration, I prefer a nice light-medium blue.

Petri Dish are fine to use for this tek right?

Not unless you want to mix up the pasty plate mix in a bottle and pour it in airbox/flowhood. That recipe is the only part of this tek that's applicable to those plates.

No this is a "no pour" tek. It gets poured in the PP5 dish before it gets PC'd. With petri dishes you PC the agar then pour in the dish.

Quick question, Something has got me confused, You use this TEK and pour it in the glad cups, Then you put the spore etc on that TEK substance in the dish right, And it grows?
Reason i'm asking this is because i see a few dish's without any TEK recipe substance underneath and just mycelium in the dish, How does it grow? Or is this because the mycelium has been transferred to a new dish and it does not need the TEK to grow/be transferd?
