Remove the gloves and get some arm sleeves is the way to go huh? Haha I've been using the lighter alcohol combo with that box for years and years! Never had a problem.. Perhaps I'm just lucky. But yeah I've seen people on here throughout the years spraying down the inside of their box with Lysol n shit then using a lighter in it.. I just have some swabs in there. Still pretty dangerous I suppose.
Anyway, just got done with the cloning. Used some inside stipe tissue, just kept splitting the step more and more for each consecutive plate to get a hopefully somewhat clean sample. Ran out of stem so I started splitting the cap. Guess we will wait and see if anything happens!

I've been wondering about this because I'm getting tired of filling my lungs with bleach or lysol. What kind of soap do folks use, and in what concentration? Ideally something that wouldn't suds up so much that the sides of the SAB are useless for seeing through.

Yeah I saw that thread. Never used ISO in my SAB.
But I don't want to use bleach anymore cause it fills my bedroom with fumes. So what kind of soap? Dishsoap? Pinesol? Does it matter? Lol

doesn't matter. don't worry about being so precise, in general. this is amateur mycology, its supposed to be simple and accessible.

I transferred from 2 plates to 8 today. Think it went well, getting the fuckin agar off my exacto knife was a bigger pain in the ass that I was expecting . The plates I was cutting were all poured to one side because they tipped over in the PC though which made it tough.
What happens if you PC plates and remove them right away from the PC? Not gonna fuck it up right?

no. I remove them immediately when the pressure reaches zero but they are still hot as hell and the agar is liquid. let them cool on the counter for nice level plates.

Yea that's why I removed them, the first time a bunch of plates were in the corner and a pain in the ass to cut out tonight. Whatever we'll see how it goooooes

Have I been wrong this whole time to think you need to flame sterilize your needles and other utensil in your SAB between jars/plates or whatever else to avoid cross contamination? Id rather have a lighter in there than alcohol swabs.. Or are you just saying don't use a lighter in combination with ISO in a SAB?
Id like to hear what box setup some of you use. Does this sound like an okay setup: disposable gloves, tyvek sleeves, inside sprayed with soapy water, lighter inside SAB?

You need to flame sterilize your needle in between inoculations if your needle touches agar. A loop or scalpel will always touch, so yes, flame sterilize in between. You don't bring your lighter into your SAB though.
I take my left hand out of the SAB to hold my flame maybe 5" away from the hole and use my right hand, inside the SAB, to stick out my syringe/scalpel/loop to sterilize it.

Lighters suck for flame sterilization. I would either buy a propane torch or an alcohol lamp. Denatured alcohol is pretty cheap.

Definitely best to flame outside the box. Other than being dangerous if you are working ISO, flames inside will stir up the the air. Ever blow out a smoke cloud and strike a lighter under it? You can see what a flame does to still air.
I don't use tyvek sleeves, just a tote with arm holes cut out sprayed with soapy water. Turn off central air/ fans. I shower and put freshly laundered clothes, gloves and mask. Everything going in is wiped down with Iso, my hands and arms as well.
