Sometimes I'll go facemask and gloves only, but wipe my arms down. I don't think Muda does much more than that sometimes, although obviously his technique is better than mine.

stareatclouds said: How much do you guys spray down your SAB before using? I usually spray Lysol on the walls, wipe them down to clean shit, and then soapy water around the sides (not the top because I can't see if I do). Then I shower and come out and use it after 20m or so. Any issues with that?
I just get a paper towel moist with alcohol, give it a good wipe down, set everything up, mist, then immediately get to work.
That's pretty much the same thing I do & it usually works out well for me. I use to use bleach water but didn't notice much difference when I switch to soap & water. Some times I still use bleach to wipe the tub down just because bleach is cheaper the rubbing alcohol.

I use rubbing alcohol, gloves, and a dust mask. I don't wear a shirt so it doesn't create extra drafts.

Very nice write up Pasty! Had agar sitting for a long time just too intimidated to venture into it. I picked up some small containers but all the had were square ones....guess they should be fine. Also the agar I bought...prob 3 years ago...is yellow. Don't know how that will affect ability to watch for contams. Going to make some of these up tonight first time with agar. Question too is why not put a RTV injection port on it to not have to open it to inoculate with the lid off? Lust curious on that...
I'm not waiting days on RTV to cure on this one. Have all I need to make these up tonight...if it doesn't work out it will still be good learning experience. but rtv port would be nice in the future if using a syringe.

Your gonna have to open the lid anyway when transferring an agar wedge to your grain jars or another plate. Just practice good sterile procedure inside your SAB, and you won't have to worry about contams.

I do like them particularly once they've gone through a few cycles and loosened up. Makes one handed lid removal a breeze. But not too much of a breeze, that would ruin the still air.

I've tried everything else. I'm sticking with Pasty Plates (tm). They just work best.

Does that mean not clean? I was planning on using them to make LI's, not isolate fully.

Nah they're clean but they're very sectored. You already said you didn't mind that though so yeah use em if you want. I always make a transfer to a new plate before I use a plate for something, so I don't actually lose a plate.

I was considering that, but am kind of in a rush with my work right now, so didn't bother. Next time, Noc, next time.
