If you have enough headspace in the top of the container, you can colonize a plate just fine. Cultures on agar require very little GE.

I don't use the containers pasty does but I keep all my petris in ziplock bags, 2-3 per bag, they have no issues :shrug:

I know you can't PC something that's completely sealed without getting the vacuum.  I guess what I was trying to ask was 'how much GE is really necessary for a culture?'  Surely a quarter inch hole with micropore tape is a little over kill?  Not saying GE is bad for cultures. But how much is really necessary when you're just talking about a little bit of myc on a tiny plate?

I found that the quarter inch was what worked for me to keep the container from being crushed in. It does not really matter if they get a bit squished anyway, just add a little extra agar to ensure complete coverage and use a smaller hole if you want to. But I would not recommend no hole. No hole is no bueno.

I made some PF cakes in the mini rounds that you do your agar in without a GE hole, what a fucking mess when I opened up the PC

Ziplock bags are not air tight.

At least they never are for me.

I always squeeze the air out before sealing, but the air always finds its way back in.

I've stored uninoculated petris in ziplock bags for a couple months with no problems. Buy the ziplock brand. The off brand are shitty

I actually put all my petris in ziplock bags. I don't use parafilm. Never had a problem

Tell ya what pussy, I'll vac-seal a bag around a dish next time I make transfers.

Obviously I won't suck all the air out, but it will definitely be an air-tight seal.

I usually do the small batch enough for 15 jars worth. If so how much honey should I add to that mix?  I Wana try yours pasty but I don't have potato flakes atm.

If your using mea I wouldn't bother with honey. Really the only reason I used PDA for this tek is that its pretty simple to make and the ingredients are easy to find. I like to rotate my media and use PDA, MEA, cornstarch agar, soak water from wheat rye or oats, and I did dog food once. Next one I wanna try is rice krispies agar :lol:

Are you using MEA powder or making the medium from scratch? If MEA powder then 20 grams per 500ml is just fine.  Never mixed it all from scratch myself though.  I did 8 plates tonight with 7 grams of MEA powder and 180 ML of water and it was just fine. You can fudge it a little.

I use 10 grams of extra light malt extract and 10 grams telephone brand powdered agar in 500 ml aka a 16.9 oz water bottles worth of water.
