When you slowly bring it to pressure do you start at medium for 10, then medium high, then high?, how slow is slow?

I get the water in the PC boiling on 8 (electric stove) put the lid on (let it vent) and drop it to 7 maybe 6 until it gets to pressure.
Stoves are different though. Once mine is up to pressure/temp, I can keep my PC idling at 15 psi with my stove set to just over the 3 Mark

Yeah I wouldn't wanna try to put the lid on my AA after it's already boiling... I guess depending on how long.

Im sure it worked fine, in still getting used to pressure cookers and over thought bringing it up slowly.lol. Got some new plates to lay some spores on. It's going to be my first time working with prints and will be going over some good tutorials I saw around here. it's late and not going to inoculate now, but excited about tomorrow

yeah i only use an exacto blade for culture work. i have 50 blades left as replacements. i used that same 1st blade for 8 years. i recently replaced it last week.

Do you think these plates from 4/20 are still salvageable? They've been in my fridge, in ziplocks with two pieces of micropore tape over the hole in the lid. I don't want to waste time cooking more up if I'm dead in the water.

If I was in a position I needed to use those I would first take a transfer to a new agar plate.
Its hard to tell from the pictures exactly how much growth those plates have though.

Were them plates refrigerated after full colonization? I'm asking because they look pretty healthy for being so old. If I leave mine out for 4-6 weeks after colonization, they shrivel and look very unhealthy. But after transfer, they become vigorous again.

I tossed them in the fridge on 5/4 so they had a couple weeks to grow out. I planned to transfer them. I thought they looked pretty healthy for as old as they were, but someone said they looked "dry" so I figured I'd ask around before jumping the gun. I was curious if I can get some jars going with whatever is leftover or if its just to old and i should wait for the transfers to grow out?

I would not drop any of those into grain jars. I would instead opt to transfer before knocking anything up, just to be on the safe side.

So i think imma try my first real attempt with agar, im not using anything fancy just some baby food jars, if i put a hole in the top nd filled with polyfil would that work out?

Glad mini rounds are cheap and easily obtainable. There is nothing wrong with using glass but with plastic you can loosen up the agar before you even open the lid and drop it into a grain jar or liquid medium without having to use any tools which eliminates atleast one possible contam vector.
