Actually it sounds like those temps would suffice. My PC says It's 250 degrees at 15 PSI.

Regular clear silicone works better than high temp RTV. It has better adhesion and any 100% silicone will withstand Temps much higher than a PC produces.

That's what I use. GE 100% Clear silicon. Wal-Mart carries it where the paint section is, by the caulking. It's good up to 400°F.

My agar all dried out! Except for one plate, mysteriously, which is the one that happened to implode slightly in the PC. Hmm. =3
Because i left it for so long in a drawer that is, i made em up in march.
So from reading i see using dried up agar is alright as long as it's clean and at least a couple of my dried up ones look contam free. Should any attempt be made to rehydrate the agar prior to use, or is it as i'd think and you'd only increase chances of dirtying the agar, introducing bacteria, what have you?
I'm gonna clean out some of the failed plates and make some fresh ones, and transfer a few chunks before i try noccing anything with agar again. I tried an LI but I think i was really rushing and my sterile was garbage so those jars turned mostly green. So before I try that again, I've been reading Violet's teks and those grass seed containers look mighty delicious and cost effective.

Is it all detrimental to add too much food colouring? I added atleast 8 or 9 drops in my last batch.. some of the growth appears discolored as if it was getting something from the agar..

I have dropped dried out plates into quart jars with no problem. But they were fully colonized before they dried out.

A couple look fully colonized to me. They're bone dry, one came loose from the floor of the mini round and it sounded like a piece of desiccated banana or something shaking around.

drop the whole chunk of dried agar into a fresh grain jar.

Is that a better move than taking bits and transferring to new plates? From a simplicity standpoint that makes sense, less handling = less chance of a failure.
I guess after I could take colonized grain to agar and g2g some fresh jars at the same time anyway.

I've been pretty stressed lately, not eating right or thinking clearly, the idea that too much food colouring came because my growth had a similar colour to my agar, just lighter

So, i made SHIP on the glad rounds (screw top) and PC'd them loose (air exchange) When the pressure on the PC went down, i let it cool a bit, removed the top, closed the inner walls of the jars, and removed them from the PC. there is still condensation on the side. Is it OK to inoculate via SHIP at this time? or did i mess it up by not waiting for condensation to go down? im worried by not waiting, and screwing the jars with condensation inside, they might contam, or am i overly cautious here? i never opened the jars.
also, 1CC per jar, right?

You didn't put a hole for gas exchange?

No. I was told with the screw tops just to leave them loose. The reason you do that with the mini rounds is pressure.

Oh, the tek uses mini rounds so I was wondering. Thanks for clarification.
The condensation will go away but if you inoculate before it does it won't affect colonization or increase contamination risk.

All my no pour plates have tons of condensation until I let them sit for a few days to clear up. 1cc is way way way way way way way way way way too much for a plate. Use one drop. I'd use less than a drop if it were possible.
