I don't know if anyone else has mentioned this yet, but I love how you can just slide the whole puddle out into your grain jar instead of messing around with cutting (unless you want to of course.)
It makes for really easy grain inoculation. Sure you could get four jars with one but it's so darn easy to just drop the whole thing in and call it good. The size of the puddle is basically the same as a 'third' of a regular petri.

It's so dry here, mine crack into pieces within a day after cutting my first wedge anyway. So there's a bunch of wedges, handy!

Sounds like they were knocking mycelium of the "freshly growing culture" that was perhaps on grains (knocked into water) and transferring them to a sterile glass vial

If you're going to dump the whole plate into the jar I would suggest these things:
1) Make sure full colonization is pretty much complete (wait a day after full colonization just in case.)
2) squeeze the bottom of the containers before you open them. This will cause the puddle to 'jump' out of it's wedge a little. From there you can just shake it around and drop it.

Meh, anna and her associate are firm supporters of vtek grows, they respond very....erm....passionatly... to anyone who questions the superiority of vtek. As you can imagine, this has lead to some very heated threads. Traditional bulk growers vrs. Vtekers.

As someone with no agar experience who's looking to get into it soon, can I ask some clarification?
You're saying it'd typically be a bad idea to let your culture grow out to the edges of the dish? Wouldn't it be better for it to be colonized than for it to be bare if contaminants are the concern? If not, how would you stop it from growing past where you want it to stay? Not sure if I fully understand.

thats when you either transfer or move to a slant and put it in a fridge,preferably not your food fridge there filthy(or you can just put the pastyplate in the fridge)

 and what Frank said (we need a :whatFranksaid: smiley) the edges of agar plate are most prone to contam because they are closest to the outside environment (spores, mold, etc) so the closer the myc gets to that environment, the more likely to contam it can be. if u don't have slants (i do not...yet) i just put them in fridge when they r grown out to this size:

that way, if a contam did enter near plates edge, u will be able to see it on the agar. if the myc grows to the edge, u may not b able to see it, and it will hitch a ride with the wedge u take into ur grain jar/plate/bag, etc.
u don't need parafilm for the pasty tek, but i still wrap them in saran wrap/cling wrap before putting them in fridge, or u can use plastic bags.

I use a 1 1/2" stir bar in a quart mason jar to stir my agar and malt extract at room temp. I turn it up as much as possible, to the point just before the stir bar gets kicked off (substantial vortex). Everything is dissolved nicely . Anybody else do this?
On second thought, perhaps it's tougher to dissolve different grades of agar. I'm using some microbiology grade agar.

I think you misunderstood me. I have microbiology grade AGAR from ebay.. My stir plate is cheap; its made from a computer fan and magnets. I think that some people who use this tek may also have a stir plate. I thought the point of this thread was making agar easier?!?! So...it takes me one minute to dissolve everything (lme and agar) in a jar vs. however many minutes it takes on the stove. Also, in this thread, there was mention of letting the agar cool back down after dissolving. Using a stir plate, could cut out a few steps. I, for one, like less steps.

After trying again it doesn't really dissolve, it just mixes well. Stove top with heat works better.

you are correct, i did misunderstand. apologies. i'm surprised to hear stirring alone is enough to make agar work. i thought heat was required, like gelatin.
for that matter, it sounds like one could put it in a blender. well anyway, a half liter of water boils quick and the pan is easier to clean.

I tried the screw-top containers for agar plates, and that didn't work out awesomely in terms of condensation. I got a LOT of it. Anyway, what I did to wipe out the condensation was turn the containers upside down and let them sit in the SAB for a minute. Then I tilted them on their sides and cracked them about a 1/4 turn to let the water drain out, then opened them gently and used individually wrapped sterile gauze to wipe out the sides and lid. I used one for every dish. This time I'm using the Pasty plate method to the letter with the mini rounds.
