I used them but I siliconed tyvek to my lids and they worked great.

Put em in the fridge. I keep em in a ziploc bag to help keep em from drying out too fast. They can last up to 6 months but its best not to push it unnecessarily so I try to use em within a month or two.

I have now done both pour and no pour agar. I like the no pour agar more as it has the advantage of:
Being more cost effective (re-usable plates!!) Easier to access tools and ingredients (Can buy this stuff in any town) Better protected against contams (No-pour bitch)

Whyte, I still wonder how you will out do yourself here in the next couple months. I am looking forward to what you will do for this community and what you have already given.

In that case you will be happy to hear that the proof of concept tests have surpassed my expectations. In fact they performed so well that it has presented a whole new challenge. I am certain the my intial prototypes will perform well but given the recent results of the concept tests my next adjustments should prove spectacular. After that some simple tests of flexibility of preperation and use will be needed however I have little doubt that they should perform as expected. Once that is complete the tek will be released

I still like to pour agar,
Petris just look nice and I honestly don't really mind doing it lol. I pour 20-25 petris a time and store them and use my pasty plates for initial germination.

Its all good. I ain't gonna advocate one over the other, they both have upsides as well as drawbacks. I have used both and will ccontinue to use both. As long as someone is using agar to ensure their cultures are clean I could care less if you use no pour or pour. I think the no pour is the foot in the agar door for most people and even if thats all it ever is my work is done

Pasty:
I have an electric PC I like to use for the small items such as this or pc'ng syringes and such. Just fast and easy to get going. It's a duo60 instantpot I believe on high it gets up to 12.5 psi So your tek calls for 40-45mins so it got me to thinking because my plates contam (before even opening). What would you suggest for a time with the 12.5 psi amount? an hour even maybe?
Oh and just a "drop" of Karo? I've been using like 2 tsps karo with 1 bar agar agar, 1 cup water and 4 tsp potato flakes. Is there a measurement of one drop?
TIA

What kind of contams? If bacteria then def try an hour. I have a pos pc that doesn't do much more than 12 psi and 45 min always works. But giver an hour and see. If it still doesn't work then something else must be wrong. If your seeing molds then I would suspect the filter as boiling temps kill molds fast.
Sounds like a lot of karo. A drop for me is like a 1/4 teaspoon. I dislike too much simple sugar in my media but a small amount is useful. Too much and you run the risk of shocking the culture and making it lazy. I would avoid the amount you are using, half of that would be lots.

I started out with a POS PC that maxed out at 11.6 PSI. I had a lot of bacteria problems until I started PCing my plates for an hour.

Hmmm.. well I use yellow agar bars... so I guess what I'm trying to describe are scattered round spots that appear a pale yellow. I haven't ever seen growth like a green mold or fuzz... it's just very thin circular pale yellowish spots that eventually take over the whole plate if I leave it.
I will try an hour and the little bit of karo. Thank you sir.

cool. I'll definitely give it more time then. Agar has been frustrating for me. Everything else I pretty much have nailed down so this should make a big difference for me as far as satisfaction with this hobby.

Don't get discouraged. Agar is the only part of this hobby that I really struggled with. I almost gave up on it in the beginning because of bacteria. I'm really glad I kept with it though. Agar has become the cornerstone of everything I do.

Good idea. I make a few sets of plate each week weather I need them or not. There's nothing more frustrating than wanting to make a transfer or take a clone only to realise you don't have any plates poured.
