So this is where I'm at now I used 6 of my 12 containers to noc 2 MS pan cyan syringe about 2 drops each, 2 Cota rica cubes from print and 2 more puerto escondido also from print. I do have a question concerning condensation.
is this to much or will the agar re-absorb it? I did the paper towell thing and when I took the foil off the towells were only lightly damp.

These are the same one's im using. Finally got it all together today. The one thing that seems really cool about these is that the agar could easily pop out and just go directly into a jar or lc with no foreign instruments at all.

Not sure about the condensation but I used a 3/8 hole with mine. Just because I didn't read the tek all the way. Hope it works out. Pcing now. 16 easily fit into my 17 qt presto. 20
seems like the max
The agar and food coloring gave an idea as well. If you did a psilo extraction with everclear and added the tincture after heating as your just about to pour. I bet you could make some hella agar doses:hehehe: In theory the everclear would act as a preservative.

I just wanted to say after all the problems ive been having with my agar mix and speed  and definition of growth.  Violet was right.  That grain water agar is the absolute nuts.  Everything grows super quick and thick with great definition........ Im feeling slightly aroused......

Yup it works good. You should see it in LC.

I wouldn't add it full strength to premixed PDA, dilute it some.

I forget who it was in the thread told me with grain water that you didn't have to mess with the potato flakes or karo/honey. maybe someone will chime in on this but I just used WBS water and plain agar. Just noced last night so I don;t know how it's gonna work yet. :shrug:

Its easy 500 ml grainwater to 10g agar.  Super definition and some really quick growth.

Pictures arent so good but this is just from, I think Friday


Which brings me to my quesion.  The first picture is P.mexicana.  Do you think I can LI with this and leave it in my 2 quart sized jars while ime away in Asia fr a month.  I wount be able to shake so any advise would be good.  I wont bw leaving for 5 days so it should have time to grow more.

How are you guys getting minimal condensation?! :/

How can I make liquid inoculation using a pasty plate? I thought I found a tek somewhere, but cannot find it. I believe it just involved squirting sterile water into the plate and vigorously mixing it and voila!

Any help on this?

Condensation inside of your agar media vessels can be kept in check by keeping the culturing area at a constant temperature of 72-75F.  Doesn't hurt to keep the RH at 50% too if you can.

You have the "grain liquid culture (GLC)" liquid inoculation technique mixed up with making an "agar slurry". 

GLC is concocted by sterilizing water, and injecting it into a jar of fully colonized grain spawn, and shaking vigourusly.  This releases the mycelium from their grain capsule and transfers the myce into the water where the myce can then be aspired into syringes and used to inoculate sterile grain mediums. 

A "slurry concoction" is how you make LI out of agar.  A "slurry concoction" is where you transfer colonized agar media into a sterilized autoclavable blender with sterilized water in it, drop your agar in the pre-sterilized water inside of the blender using the wedge and transfer technique, and blend the mix up.

I don't worry about condensation. It clears up 1-2 days after the PC cycle.

I believe he is refferring to my Easy LI video. Make sure you don't use too much water with this tek. 1 pint is about the limit.



If you have blender, you can also use that a bit more effectively.

1. Bring your PC up to psi as slowly as you can, and then bring it down as slow as you can too.  It's a pain in the ass, but I just cut it back a little bit at a time over the course of an hour or so, then when it's off let it cool naturally.  The more patient I am the better my plates turn out


2.  Absolutely.  Drop a plate in some sterile water and grind that shit up.  There are lots of methods.  Munch made a simple one in his mushroom cultivation videos series.  Also Muda has a great LI tek with other ways to do it.  The world is your King Oyster
