So made some new pasty plates last night, this time I actually measured out (weighed) the amount of agar and potato powder and I sure was using WAAAAY too much!
So today I dropped some spores on the agar and fingers crossed I can actually get them to germinate! The agar looks a lot better as well, I think I might be getting the hang of this. At last.... Well, actually. I just think that it is the fact that I weighed it out.

So my pressure cooker is a dud! well it only pressurises to 10psi/70kpa.... is this sufficent to sterilise agar plates?
or can i just sterilise Agar plates in an oven or standard pot with lid?

10psi is better than a pot, just run a it little longer and it should be fine. I actually gave the 45 min benchmark with people like you in mind. If your paranoid try an hour but I am sure 45 min will be fine for you

after sterilised agar plates, then transfering either spores, flesh or liquid spores, whats the best position for them to sit while waiting for the Myc to grow?
do they need to be dark? or with regular light like a day cycle?
sorry, i know im a noob and am asking s*%t loads of questions, but i like to be very clear and i am documenting a grow for extra noobs in lamans terms

I leave my on top of my speakers so they can get some natural light.
Also allows the myc to jam to some good music I find the music helps with growth, the type of music effects how they'll turn out :P

Ambiant light on a 12/12 schedule is good during all stages of development. I usually just put the somewhere out of the way, where they won't get dust rained down on them.

Anywhere your missus doesnt give you that look and say 'You doing it again, its either me or the mushrooms.'
I have been using the airing cupboard recently as the temp is perfect but it is only dark in there. Im not sure what im losing incubating in the dark. I also have a shit load of stuff on a shelf. Temps between 60 to 80 work, the hotter the quicker growth but this also encorages contams to grow.

My agar mix doesnt liquify like the pictures. However, I spooned it all into the minI rounds. Was I supposed to just use the liquid or all of it?

Your agar mix doesn't liquify?
What does that mean? You mean when you put it on your stove? Let it sit there for a little longer, it will dissolve eventually but not completely. Then you spoon it into your mini-rounds when it's all pretty much the same consistency and PC.
If you have big chunks of agar or a bunch of un-dissolved potato flakes sitting in a corner your agar won't solidify/you won't get the same balance of nutes.

Put it on your stove on low until it dissolves. Add the water, agar and potato flakes and wait for it to dissolve, stirring occasionally to make sure it's spread out. Then add a drop of karo.

PC for 40 min. Bring pressure up very slow. Let PC cool over night (thats how I have always rolled). Night PC morning sterile work. Oh, an injection port for the lid.
Thanks Whyte.

Thought I was gonna fuck it up, but i've gone some nice red discs o' agar happily being colonized.
It's strange how some mycelium grows mountainous, like a half an inch high and thick looking while some are just barely coating the surface it looks like. Is this a sign of anything other than genetics? disproportionate nutrient distribution? Weird.
I know this is a slight departure but would putting a stir bar (portion of a metal hangar coated in silicone) in the bottom of a jelly jar and then adding a thin agar layer and pcing and then inject with ms or lc or whatever you got and let colonize. squirt in some sterile water and use the stir bar to sort of blend with my ghetto pc fan stir plate? keeping it all clean without opening the jar, you'd have some nice LI right? if the stir bar could make it through the agar+mycelium.

I am gonna give these a try, but I am implementing a siliconed on tyvek filter instead of micro-pore tape.
This decision to try this tek was caused by me moving to hastly in my still air box and blowing some green mold spores onto my half pint dishes.
I like the idea of being able to work with it upside down.

If you are going to attach a filter with silicone you may want to use something that holds up better than tyvek.
