If you're asking whether I want to be happy or unhappy then obviously I want to be happy.
Taking away those two conditions though, I think I would rather have a huge knowledge base on a few subjects. Assuming that it doesn't cripple my ability to live life like a normal person.

In this scenario you would be significantly less happy than the zen master but not necessarily unhappy, just less satisfied with life altogether until you achieve your lifelong achievement/breakthrough at any given time.
the catch is you don't really know when/if your knowledge will pay off and the longer is does not, the more unhappy you become

I'd never want to lose my ability to understand simple things. I feel like much of "human achievement" that everyone gets so excited about is the result of inflated egos. Overachievers who focus too much on one thing tend to lose focus of the big picture and cause more problems than they're worth in my mind.
I do often wish I had more focus rather than being all over the place with my interests, but thats just not me.

When you phrase it as "Unhappy with an eventual breakthrough//Happy with no large contributions" than, to me, the choice is obviously to be happy--but the question you are asking isn't so much what type of knowledge do I most desire, as it is a question of what type of value I place on both happiness and fame/wealth (assuming this is what a breakthrough would bring).
Removing the happy/unhappy context, my answer would change--the way I see it, knowing a little about everything essentially amounts to useless knowledge--you may preform well on Jeopardy, but your knowledge doesn't really transfer from the realm of trivia to that of practical. You know who invented the car, but you can't build one yourself. I believe it would be more valuable to be highly knowledgeable in a few areas to the point that you are able to apply this knowledge practically and with a high degrees of skill--and therefore, I would rather know a lot about a few things than a little about everything.

People care if you make a breakthrough. People don't give a shit if your happy. For the good of mankind, I would pick to be unhappy.
In real life, I prefer to know a lot about a lot of things, be happy and not contribute shit.
