Did this somewhat successfully a few years ago.  This time around, starting a couple months ago, I started a few lc's.  All grew quickly, but all were contam'd when I put them to wbs.  Cleaned up my act a bit and tried again with each lc under much more sterile conditions.  All went bad.  All were super quick too and from what I've read, the quicker they are, the more likely your lc is bad. 

So, started over about 8 or 9 days ago.  Had 2 10cc vials.  Used half of each in cakes and put the last 4 to 5 cc's into two separate lc's.  Light karo, 4% karo to tap water in a 1/2pt jar.  Growth in the brf cakes is progressing nicely and clean!  Happy day, fixed one issue.  But the lc's are barely showing any growth.  Tiny tiny strands here and there.  Like a 500,000/1 ratio if I had to put a rough estimate to it.  Followed a trusted tek I found on here for making the lc's so I'm wondering if I used too much ms solution and threw off the ratio some how?  1/2 pt jar 3/4 full had 4-5 cc's of a spore rich solution added.

Any guesses?

Stop using LC and move in over to agar. Less contams and easier to work with with much better results.

Spore syringes are almost never 100% clean so any little contam is going to grow in a LC like mad. At least in a PF cake it has a chance not to manifest so you get away with syringes. Starting a LC with a syringe is a sure fire way to lots of failure in your future. And then if you want to start a LC with a syringe a single drop of spore syringe solution is enough same with pf cakes 0.25cc per hole or less. I would just skip LC all together. The syringes are faster than letting spores grow in a LC only to inject a PF cake and then wait for that to grow all the while spores would have beaten you out on time. You have a better chance of making another clean spore syringe from the fruits you see than making a clean LC from spores so just make more spore syringes until you want to get into agar work.

The cakes are doing well.  Seem to be getting quicker as more and more rhizomorphic arms stretch out.  Cool to watch this stuff do it's thing.

Watching the green mold grow in my last couple lc's was quite entertaining until I realized it was mold that I was growing.  lol...  That shit grew fast.  Lost a few bucks messing around with it but learned a bit too so it's all good.

I never removed the vacuum from the jars.  Will that effect growth at all?  Was just browsing mycopath and saw that they suggest introducing fresh oxygen and releasing the vacuum from pc'ing.

May as well try to keep them going as they have $15 in spores floating around in them.  lol...
