I still suggest doing a LC rather than G2G, you have already seen how slow agar-grain can be. If you do a good LC you can nock up enough jars to do your whole project, or at least whip up one jar of LC with a self healing lid(silicone) and nock it up to see if you like it. It is faster than MS inoculations and MS is obviously faster than agar-grain.
Although with G2G the receiving jars are quite fast, the time it takes to go from agar-LC-grain is faster than agar-grain-G2G

I'm never using a needle again. I only need to worry about GE on my jar lids.
Have you seen this yet?

Doesn't hurt to try it out, having a source of a good LC isolate will get you where you need to be, then you can put you SAB away until you have another isolate to make another LC with.

I'm already where I need to be. Things are going good I have plenty of petris and grain jars going just fine, I just wanted to know about this batch and why I was seeing what I was seeing. You're
You trying to convince me to use a LC is akin to trying to make a straight guy suck a dick. It's not going to happen.

I can't argue if it works for you it works, but I do see it as inferior technique. I would rather not cut corners and just find out what's the real root problem and it seems to be trying to get an isolate from clone tissue especially being grown out on grain then expanded to coir then the fruit went to get 3 transfers on dishes has likely made it too tired to work quickly.
I have a bunch of knots in my substrate so pins in a few days and fruits in a few more. I'll just use prints from those to agar and start fresh again.

bodhisatta said: Ok so my revised question list is
1. Clone tissue from a MS grow is sub optimal to obtain an isolate from? if you use clone tissue from MS the best practice to use it for is take one transfer to a new dish and use that whole next dish to inoculate grain.
2. You think for the reasons in question no1 I'm seeing poor performance from these agar wedges?
3. If you want an isolate you should start from a very small(as small as possible) amount of spores onto agar. Then try out the isolates after they're obtained to see their fruiting characteristics?
1. CAN be sub optimal, but not always. This is not a stead-fast rule, but it does happen.
2. That's what I think. But your grains seem a little dry too. Since they do not seem to making progress, a good shake may be in order.
3. Yes. Test all the isolates to see which ones you like, not just the fastest. The fastest one may not even fruit. But, it's a matter of preference as to whether the slower colonizers are worth testing.

It felt weird but I did give the jars a small shake to break up the little ball that had formed so the top is now covered in more inoculation points. I have some other jars I put wedges in a few days after these ones in question that are already further along so I won't be too sad if these don't go well.
It's hard to know how wet the grains are going to be after the PC run so I always try to error on the dry side. My drying process after the boil involves transferring from colander to colander slowly. As the grains pour from one into the other they let off steam after maybe 10 transfers back and forth they easily pass the toilet paper test and that takes maybe all of 5 minutes to do.

Post your results of shaking. My thinking on this is the top may be drier than the rest.

All the jars are doing wonderful now. All it took was a little shake to get more points around there real close to being about 1/3 done now so a good shake is in order soon.
I guess I just need to use large wedges.

I've already spawned half of the batch of those questionable jars in my mini-brick setups and the others are at about 90% now.
I do 1q of spawn to 1-2q of coir in a little plastic shoebox and then fruit them in a SGFC.
