Agar to Agar transfers don't always take hold
    #19154110 - 11/18/13 01:19 PM (10 years, 2 months ago)

When doing isolation work, sometimes the transfer to a new dish won't recover and take hold, and then it's wasted genetics and petri dishes.  Is there a secret to getting a higher success rate?

I've only very rarely seen this happen and it happened to me only when my agar mix ratios were off.

I'd recommend stirring the agar bottle when pouring, every 3-4 plates I give it a good swirl. This prevents the ingredients from settling, and solved my issue with what you are describing.

Can you post pics?

make more than one transfer on each dish or make more than one transfer of each section to separate dishes that way if one transfer shits out then you have a back up of the growth you selected.

Using the PDYA recipe from your agar thread Frank with tap water and getting excellent, rhizomorphic growth from most of the transfers when they take hold.  Lab grade agar powder, flaked nutritional yeast, pure potato starch and dextrose from the brew shop.  Been giving it a whirl every few pours as well.  It's just the one here or there that isn't recovering at all after the transfer it'll just be a sad little agar piece sitting alone in the middle while the rest in the stack are going along just fine.  Like can you imagine being that agar piece watching your peers grow and move on to better things while you got held back and never did anything with your life, before being tossed into the trash by a frustrated cultivator who had such high expectations of you, only to be let down?  How depressing.
