How to view clamp connections
    #19149097 - 11/17/13 10:38 AM (10 years, 2 months ago)

Can anyone that does microscopy work explain how to prepare a sample to view clamp connections and other fungi characteristics under a microscope?

The simplest method I use for viewing is to have a clean slide ready and then take a piece of clear tape and gently touch your petri dish or other mycelium with the sticky side, and then place it on your microscope slide.  This holds the mycelium in a flat plane so it stays in focus.  Obviously this is inferior to mounting a proper slide but it can be done in seconds.

To see clamp connections, slowly raise and lower the stage to change the focus point as you look at intersections until you find one.  The mycelium will get 'fatter' in that spot.  You'll probably have best luck finding them with 400X.
