Just inoculated my first PF Tek jars and I have some Q's

Alright so last night I finally got up the courage and had all the materials to start. Used the classic recipe of verm BRF and water. Tried to pressure cook them but pressure cooker was garbage and wouldn't hold pressure I think the rubber gasket is done for. So I boiled water with them in a pot with the lid on for 90 minutes, adding more water at the 60 minute mark. I let it cool over night without opening the lid.

So this morning I went to inoculate them. First off I took a shower to get rid of any spores bacteria and dead skin.  I took the pot downstairs to a bedroom which I figured was one of the more sterile places in the house. I Ousted the room pretty well. I then took my SAB that I made which is just a bin with two holes I melted in it and washed it inside and out with a soap and water paper towel and left it. I used antibacterial wipes on my desk and put the SAB on top of it. I put on my powderless nitrile gloves and removed the lid of the pot. I then washed the gloves with 70% iso. I then removed each of the 5 jars and put them inside and closed the lid on the SAB. I took my syringe and removed the plastic covering and washed it with a tiny bit of iso still left on my gloves. I then removed the needle from the plastic packaging and put it on the syringe. I think this is the point where I remembered to put on my dust mask unfortunately I didn't do this sooner. I shook the syringe a bit and put it inside the SAB after flame sterilizing most of the needle without getting too close to the plastic part as to not melt it(did this outside SAB). I took the foil off the first jar and attempted to inoc it. The syringe was overfilled and so as I was injecting the jar I couldn't tell how much solution was actually going into it. I took it back out of the SAB and squirted a bit of air out of it and added a tiny bit more to each of the four holes. I didn't have many issues with innoc after that I tried to eyeball 1/4 cc for each hole and ended up with 6 cc left in the 10 cc syringe but I estimate there was about 1 cc extra in it.

I flame sterilized outside of the SAB between each jar. The holes were drilled with a 1/8 inch drill bit and the tops of the jars were inverted so the rubber seal is up.


I tried to get the needle off and put the little cap back in but it was on super tight. Do you guys normally leave the needle on for next time? I flamed it then put the needle cap back on I hope this is okay.

Also how important is shaking the syringe? I shook it twice or three times for the five jars I believe. The ball of spores didn't want to break up very easily and I was afraid I would do more damage by stirring up the air in the room.

I also inserted the needle most of the way in as to make it past the dry verm layer so contam wouldnt follow the wetness downward. However it was hard to get the base of the needle red hot as I didn't want to melt the seal.

I plan on leaving the foil off and there is no tape. Just the dry verm layer as protection.


Sorry this is long winded I thought I had the basic process figured out but actually doing it gave me a lot more questions than expected.  Any tips on my sterile techniques or anything else would be much appreciated.


EDIT: I just ousted my closet and used sanitizing wipes right in the immediate area of the shelf and sat them there.

Sounds like you did you're research and took just about EVERY precaution.  Id say you have absolutely nothing to worry about.  But don't get discouraged if a jar or two does get contaminated.  It's just part of the hobby.

Now that the jars are inoculated.. don't touch them.  You don't want to disturb the verm layer.

But anyways. CONGRATS on starting your first grow =)

Oh and about shaking the syringe, I'm sure it was fine since you could actually see the spores in it.  Shaking just helps.

Thanks I appreciate the input. Feels good to finally get some jars inoculated. I've been browsing the cultivation sub forum for years. I finally felt it was time and things just came together. :grin: If I get at least a few beauties and get to watch them mature I will be happy.
