My First Grow PF tek BRF
    #19095135 - 11/06/13 12:27 PM (10 years, 3 months ago)

Wanted to document our first mush grow and hopefully get some tips from all the knowledgeable people on here.  I've been lurking for months, and reading as much info as i could before i started.  Now that my brain is completely saturated with info, it's time to get some practical skills.

Right into it...
We mixed up the 2 parts Verm, 1 part BRF and 1 part water mixture based on the pftek method so widely used on this site.

After mixing it up, the consistency and moisture seemed to be pretty good....was moist, but when i squeezed it, no water came out.  Filled up 6 jars..well 2 jars, 4 glasses.  We only used 2 jars because even though they say "widemouth" there seems to be a little indent on the inside that might keep the cake from coming out.

After filling all the jars, we wiped the top of the glasses clean and topped them off with dry verm., covered with tinfoil, then off to the stove.

We boiled them for 1 hour and 40 minutes....a little longer than called for because we had to add a little extra water to it to keep it from boiling dry.

While we waited for the jars to cool, we build the FC and an inoculation chamber as well.

Once the jars were cool, and the chambers built, we wiped down the entire area with lysol wipes, and the inside of both chambers before we placed all the things we would need in it.  I was really worried about contams so i wiped the packages of the wipes and spore syringes before they went in as well.  I then used an alcohol wipe to do the gloves before i started the inoculation.

After flaming the needle till red hot, i wiped it clean with an alcohol wipe to remove the soot.

I then proceeded to inoculate the jars.  We used two different types of spores...blue meanie and john allen....not for any other reason than we wanted to see which was more forgiving.
I gotta say, this was a lot harder than it looks...for me anyway.
I have a question about this part....when i inoculated some of the jars, there was a little spore solution coming out of the tip of the needle without any pressure...when i inserted the needle, the tip was already wet..is that ok?
I flamed the needle between each jar, and alcohol swabbed right after.

after all the jars were done, i placed them in the fruiting chamber till some mycelium grows.  Do they have to be in the dark?  The ambient temp in the FC is around 23C.  That's where we're at so far.  Now we wait and keep reading!!  Thanks for any help you fine people might have to offer.

no need to keep jars in the dark. But also useless to wipe the needle AFTER flaming it. Flaming the needle till red hot is what you need to do. Do not wipe with alcohol after you flame the needle.

will the black soot left on the needle not hurt the inoculation?  They're not really in the dark, there's some ambient light in the room...it's just the pic...Thanks for the reply

apparently there are mixed reviews on whether to flame or not, or whether to wipe or not.  Some say they don't flame, and some say they don't wipe, and both sides claim to not have any major problems, so by doing both, i guess i increase my chances of mycelium growth.

These forums are endless....all the reading i've done and still have a million questions...

make sure your checcking for the right things, some say they have no ssues with wiping but for the most part there doinng  grains hich tthere needle ti will nver touch, and it doesn't get any more sterile then red hot, the first drop of solution will cool your needle, you can wipe before you flame np(poitless but u can)

I only just finished my first flush of my first grow, but, it looks as if you're following the tek.  My only suggestion would be to get the jars so they're all consistent, but I guess that's not vital.  Good luck, you'll have fun watching them grow.  Prepare to be amazed with the first big growth spurt.

Chemicals sanitize, heat sterilizes....there are no mixed reviews.

Never wipe the needle, ever, not even when putting it away for storage.  It may look ugly and black from soot but if that bothers you you can buy replacement needles at your local pharmacy.  Just don't go in looking like a junky.

I've never understood why the soot bothers people so much when they KNOW it's sterile.

I have a question....when i inoculated the jars, some of the fluid came out of the needle (without any pressure on the back of the syringe), so when i pushed the needle through the dry layer of verm, would it have dampened the dry verm layer enough to invite contams?  It was my first time inoculating anything with a syringe, and was a fuck of a lot harder than it looks.  I appreciate your patience with me, i know i'm  a noob...any feedback would be great.  Spanks :syringe::tongue:

make sure your needle is tight to your syringe, you will probly be fine just try not to pick the jars up that much so you don't disturb your verm barrier

So when i woke up this morning...this is what I saw.  We inoculated them on the 6th.  I've seen no mycelium growth yet.  The temps were low at first...around 21-21 (68-70) but for the last 3 days they've been up around 24-26 (75-78).  I've read on here that it could be lipstick mold, or rust.  It is also in the other jars, but only where the inoc holes are, and it appears as though it's only where the solution is present.

Can anyone help?  I can post more pics if needed.

Just wait until further examination is possible, it could be two weeks before any mycelium growth is present.

Thanks!  We knocked up another 4 jars with Golden Teacher tonight.  We've got room in the FC and it'll save us a week if the first batch turns out to be contaminated....got my fingers crossed!

Yesterday when i checked on the first batch of jars, i saw this at the bottom of one.  I thought it strange that it appeared at the bottom, but if you look at the orange colored substrate just above it, that was the first one i knocked up, and put way too much in it....so the spore solution went right to the bottom.  For some reason, every jar i knocked up, the spore solution appears an orangeish color.  IS this contaminated, or the beginning of mycelium growth?

thanks for all your help!!

hey po. nice glove box!

Quote:

yes, that should be fine... as long as you sterilized the tip of the needle properly, which sounds like you did though if the solution was exposed to the outside it could contaminate the jars.. but this is not as likely considering you are using a glovebox.

Quote:

they dont have to be in the dark, but light is a pinning trigger. IME it's not really a huge factor in things, even when doing larger substrates. the largest factor for pinning is really the evaporation of the substrate, though co2 content and fresh air exchange FAE play big parts too.

you should keep the PF jars in room temperature.

when these colonize, what have you chosen for a fruiting chamber? i would advise against a SGFC from experience. a hybrid system would be better suited for cakes, one that utilizes a monotub-like design with perlite at the bottom.

Thanks StateOfMind404!!  I went with the SGFC because I'm following the BRF tek to a T....at least as closely as i can.  What problems did you have with he SGFC?  What would you do with a SG to modify it?

i had issues keeping the humidty up in the SGFC, though there are vets that will tell you that if you do it properly it works fine, though i did things like i was told and it was still a pain.

Doc-T has a hyprid system that takes the good parts of a monotub design, and if i was utilizing the pf tek i would build myself a monotub-like chamber... basically has wet perlite on the bottom, and holes filled with polyfill or micropore tape at the bottom, and holes at the top to create air flow.

the wet perlite is to elevate the humidity in the chamber. the holes should be 1.5 to 2 inch holes i believe. those are the only holes you will need, instead of a ton of little holes.

Would it retain more humidity if i covered every other hole with micropore tape...or stuffed them with synth stuffing?  I read another members thread/tek where he made a FC but used only the larger holes filled with the synth stuffing.  Put a fan on in the room to get some airflow, not directly on the FC and it looked like it was working for him.

I'm still so new to it all I'm just trying to get my feet wet, but if I do anything, i like to do it right.  There are so many methods for cultivating these beauties, I'm sure i'll get one sooner or later.

thanks again to all the great minds for the wealth of knowledge on this forum!!

I saw this today on the same jar that has the myc growth on the bottom.  The inoc holes on the top of this jar are a little larger than all the rest, cuz it was the first one i did, and was all shaky, but i think the larger holes are helping the GE, so i flame sterilized a knife tip and made the holes on the other jars slightly larger to see if that might speed up the myc growth.

The orange stain around it is where the spore solution was distributed...i dunno why it turned orange, but i'm gonna wait it out before i toss'em.

Cheers
