recently streaked some amazonian spores to agar from a spore print i made 1.5 years ago. NOTHIN' Sterile? non-viable? have had good luck with this strain in the past. The spore print was very dark and these suckers made a ton of spores. any thoughts? yes, i cooled the inoculation loop in the agar before i streaked the spores so that aint it. here is a picture:

I'd guess it's the age of the print.
Try again on a few more dishes and see if you can get anything.
How long have your dishes been inoculated?

Try hydrating them in some distilled water for a few days first, if that fails and your desperate try longer. Failing that you can find a way to plug the end of the syringe and use elastic bands to apply pressure to the plunger with the theory that the extra pressure will force water molecules into the spores. Keep it under pressure like this for a few days. If that fails I would give up hope.

A lot of people have been having this problem lately....I think it has to do with the spores not having enough moisture to germinate.
Did you apply water to them after placing them on the dish?
Was the agar really dry?

Hehe, great minds and all that
I tried it last year with some v.old z-strain spores but sadly wasn't able to get them to germinate in the end, they must of just been tooooo old.
If your struggling with ideas of what to block the needle with, I melted candle wax into the little plastic bit that goes over the end of the needle, then just pushed it back onto the syringe forcing a small amount of molten wax into the very end of the needle, which set and then blocked it.
Worked quite well actually, shame my spores were just too far gone.

Thanks pal. I will let you know. I will try some water first. Gonna be sad if I lose this one.
