any point in agar work without hepa flow hood?
    #19089402 - 11/05/13 10:31 AM (10 years, 3 months ago)

is there any point in attempting agar work in a still air box i will be using cling film instead of parafilm and then putting them in bagies or is this idea pointless waist of time. i know the cling film part works done it when i had a hepa but are the bagies overkill.
before people say use search at least let me know what terms to use
thank you

Use a SAB. Lots of people do it all the time. I do.

The baggies are not overkill, in my opinion. I use parafilm and sometimes still use baggies.

Agar works just find in a still air box - but expect plenty of condensation on the inside of the box. Make sure the top of the box is slanted so that water doesn't drip into your work.

Parafilm is also just fine. It breathes less, but I've used it many times with success. You can use a sharp knife to chop of an inch of the roll, the perfect width for wrapping plates. I do find I get more contams with parafilm (I assume to to very small gaps between the disc and the film?). If you can tolerate a few contams here and there, don't worry about the baggie.

I use a SAB while I'm very :blazed:

Sometimes this fucks with my sterile procedure but I get quite good success with dishes even when I'm in a hurry. If you take your time and do premeditated choreographed moments you'll do amazing.

SABs are indeed a fraction of the cost, but I think the sterile airflow makes for incredibly reduced hassle, and keeps the environment cleaner as a plus.
IMO the cost of a sterile airflow cabinet is nothing compared to ease of convenience & success it affords.

O.P., consider using containers for your agar dishes - reduced failure rate, cost, effort, hassle, and materials

I use a SAB to put spores on agar and to do G2G transfers - it works great for me.
