LC test jar
    #19079007 - 11/03/13 06:47 AM (10 years, 2 months ago)

So I've never really dealt with LC's, when we test a jar is it imperative to fruit it or is it enough to simply let it colonize contam free? If you do need to fruit it, I need some suggestions on how to fruit a quart of spawn with no GH.

Obviously crumble and case would bs my main option.

Seeing that it colonizes contam free would seem to suffice but like you I am inexperienced in the ways of the lc. Hopefully someone will chime in with some experience. I'm getting ready to make a galindoi lc from an agar wedge. It will be my very first lc

I'm planning on give LC a shot this week sometime for my galindoi, mex A, and pan cambos. Mostly just concerned about my lack of a stir plate and if swirling a marble around in the jar would really be enough for its O2 needs.

I do a quart of spawn to 2 quarts sub in a tray all the time for testing clones, ms, etc. SGFC is a good route. These were both 1:2 tests;

The thing that sucks with LC is all the needle aspirations they go through, and wiping the SHIP with iso doesn't sterilize it, it just sanitizes it. You are chancing pushing some sort of contam through every time you stick the SHIP with a needle, which has to happen quite a few times to aspirate all the LC into every syinge. The thing is, it will contam EVERY jar if it does go bad. Thats why you gotta do a tester jar first, but even just the act of aspirating some into a tester jar, could contaminate the LC so anything inocd after that is kinda always a "tester" if ya know what i mean. And if your filter gets wet, it can easily invite bacteria directly through it into your LC.

Im just naming all the reasons RR and other experienced cultivators talk down on LC. They do work, but with a lot more areas that can get contams. I myself used GLC a lot but had some contam problems here n there so im gonna move on to g2g and agar. I'll use LC when i get a stir plate, or for species that dont take kindly to a shake at 35%.

Agreed. This is why I'm not using LC for cubensis, (an unneeded risk for something you can shake to expand) and why I will be using 60cc syringes, which will hopefully cut down the number of times I need to stick the SHIP. Little details that go unheeded too often which is why I think so many fails are reported with LC. I would never advocate use of an LC to anyone who does not have lots of experience with agar first, to learn proper sterile technique, and is why I waited until now to start experimenting with it (that and mex a is just to slow to nock with wedges)

And back to the op. When you use a test jar, is it enough to let it colonize or do you need to fruit it for some reason?

Like stated above you just wanna see clean growth unless its an iso that hasn't been tested yet in which case I would fruit it to see what I got before knocking up a bunch of jars

Shot glasses filled with brf ala PF tek are really fast ways to test your LC. Or even buy a 8pack of 1/4 pint PP ziplock jars for 2$ and use those as cakes to quickly test LC.
