Mycelium Stunted: Next Steps?
    #19074863 - 11/02/13 12:16 PM (10 years, 3 months ago)

Hey Guys,

I wanted to get your opinion on how best to move forward here.

I inoculated 12 jars on Sept 16. with Golden Teacher - it's been 47 days, and out of the 12, 5 showed no signs of growth what-so-ever, so I tossed 'em. Of the 7, 2 made it to full colonization, while the remaining 5 seem to be taking quite a long time. Throughout the process I've noticed these guys really having a tough time from the first signs of growth and it has been about 2 weeks since it seemed like these 5 jars have come to almost a complete stop. These jars have been kept at a fairly steady 79-84 degrees throughout incubation.

I wanted to know at this point in the process what the best route to take is with these guys? Over night in the fridge? Just don't touch 'em? See if I can salvage the mycelium and grow straight from jar?

Right now there appears to be no signs of contamination at all, and I wanna make the right moves as I feel like that may become an issue the longer these guys sit with no signs of growth.

Maybe the cakes are to dry?
Do you have fresh air exchange? cant see on the pics.

Use a marker to draw a line on the edge of the myclium, wait for a couple
more days, so you can check if they really stopped growing, sometimes very hard to see

The inside of the jars look wet as hell. Water spots everywhere. Clearly doesn't let me know how wet or dry the inside of the cake is, but I have assumed the water conditions have been OK.

Gas exchange could be an issue, but the jars do have the 4 open holes on top, so unless I open 'em up, I think they should be alright in that department as well.

The marker idea is brilliant, so I'll do that now. Also going to toss these guys in the fridge for 24 hours, as I read that technique can sometimes help.

Is anyone familiar with Golden Teacher? I do know some strains are slower than others, but either way, 47 days and less than half way along seems too slow.

What is temp of your incubation room?
Golden teacher should't be that slow.
I once tried Golden teacher, all jars where fully colonised within 4 weeks.

  The name some vendor gave some group of spores is irrelevant.

All those jars look contaminated....none look healthy...to me anyways.

They are also upside down for some reason....which is never the right move.

Also, this looks like a kit, which explains a lot.

I agree with hacker. Contam and why the upside down?! That hardly allows gas exchange (among other things).

To me it looks like bacterial contam.  Look at the edges of growth.  Are they wet looking? or wispy?

Don't put them in the fridge either.  I don't know where you got that idea but its not going to help.

Do they smell stinky?

I'd hit it.

No, they smell delicious. 2 of the cakes finished. Really nice smell to them.

I decided to sacrifice the cake that was the least furthest along to have a closer look. The smell is fine. Not wet on the edges, but definitely more webby along the edges than any of the other cakes. Here is a close up. Am I dealing with contams?

Here is a shot of one of the jars further along and doesn't seem to resemble the slightly webby thing that's happening on the cake above.

Thats a good sign!

but honestly, I read so many contradicting things regarding growing, it's damn hard to isolate truth from fiction

Unfortunately, thats the way it is. Look for the trusted cultivator tag when in doubt.  (RR, anno, Frank, etc etc) The search function has an option for only trusted cultivators.

What kind of contamination are you guys seeing? The color looks total fine to me with just a touch of mild yellowing from water spots. No browns, greens, or even noticeable webbing.

Is there something you guys are seeing that I should be looking a little closer at? If you tell me what you're seeing I can take a closer look.

Potential signs of bacterial contamination include wet blotching, weird smells, stalled growth, lots of metabolites being excreted by the mycellium, a wet look at the edges of new growth (rather than wispy or rhizo-like), slimy or soft spots, etc.

The air exchange should ideally be at the highest points of the jar or bag, because the heat emitted via respiration of the fungus creates air movement up and out.

Hey Joiningheads,

I added detailed images to my post above. Would you be willing to take a closer look and let me know your opinion on contamination?
