First timer and extremely nervous about inoculation

I am using a PF Tek grow and I inoculated the jars on Monday, but I gravely misread instructions on inoculation and I injected the spores straight into the dry vermiculite. The thing is, I placed them all in about the same area (which I'm hoping dampened the area enough to allow for spores to start growing) and now I'm panicking and thinking I just screwed up my first grow. Is there A) any chance that they will grow, but the cake will be delayed or B) any way in hell that I can salvage this. I know it's still too early to tell, but it's eating me up inside. Thanks you guys.

I guess if the spores find their way to the wet substrate leaking through the dry vermiculite then they will germinate but colonisation will be very slow because it starts from the very top and not the center.

At this point I would be more than happy to wait a few more weeks just so long as they germinate. I don't know if this makes any difference, but I am working with Great White Monster spores

It's not the spores germinating that you need to be most worried about. By wetting the dry verm layer you just gave contams a direct route into yr cake buddy :thumbup:

Hey OP,

I am on my first grow here and one of my jars has an extra thick layer of dry verm (due to running a little short on BRF/verm mix).

I did not realize that my syringe would then not reach past my dry verm layer. It was the last jar so I injected it any ways.



This is day 11 and first signs of life are starting to show. 1 jar at full colonization. 2 around 85% and others around 50 to 70%.

give it some time I'm sure something will pop up.

Even if you did inject it into the verm, the spores will probably find their way down onto the BRF/Verm.  Trich does if a sterilized jar is left long enough.  Spores are tiny and light so they can keep moving until they find something to grow on.

I have been keeping an eye on the jars for the past couple weeks and I haven't seen any growth. I'm gonna check one more time when I get back, but it looks like I did screw it up :frown:

okay a little bit of news to dampen the blow - I found that three of my jars are actually colonizing! One is really early state (I believe I saw it collecting in the middle of the jar), one has completely colonized the topside of the jar, and one has almost gotten the entire jar. I had originally 18 jars going so this wasn't a huge success, but, dammit, it's something :grin:

Wait, what? I thought I had to keep the foil on them. I've kept the foil on all the jars the entire time. Do you think that has something to do with the slow growth?

Foil gets removed after sterilization and stays off.

It should not hinder GE, but it could moisten the dry verm layer, which is not good.

This is why it is recommended to keep the foil off, so the dry verm layer can stay dry.

I just removed all the foil off of them. The two jars that currently have a sizable grow have the top layer of the vermiculite being consumed into the cake because of an error during inoculation.there isn't anything protecting them now and i'm sorta scared

I've always heard people say leave the foil off for the GE not to prevent dry verm layer from getting wet. This makes a lot more sense thanks Nota. :thumbup:

I think hes right.

Could be it some sort of visual illusion, is there
hard water buildup on the bottom half of the jar.

Before you know for sure, I'd isolate that jar, and any
that look like it.

Take a picture in decent lighting, or with flash, and
post right away.

I had something similar that looked like that from the steam sterilization process.  It was only on the outside and was fully cleanable with some alcohol.  Hard to tell from the pic tho if its the hard water buildup on the outside or an actual contamination.

I took the jar out of the incubator and set it away in a corner of my closet that's dark and isolated. Here's a link to some pictures I took in better lighting

Jars/bags/tubs/trays should colonize @ room temperature getting ambient/indirect light.

Main pinning triggers are full colonization, FAE and Evaporation off of the substrate.

Light is a secondary pinning trigger. For tropical species temperature is not a pinning factor.

P. Cubensis are a tropical species. You could colonize at 70F and fruit at 80F with great results.

Light has been proven beneficial during all stages of mycellium growth. Mushrooms like mammals have a circadian rhythm.

You want ambient/indirect light(on a 12/12 schedule preferably) for colonization and consolidation.

You want direct/intense 6500K light on a 12/12 schedule for fruiting.

Optimal temps are mid 70s throughout the whole grow, but anywhere from 65F-80F is acceptable.

Incubation is outdated/uneeded unless temps in the range stated above cannot be kept.

The inside of the jar is always a few degrees warmer than the outside because the mycellium produces heat..mycellium tends to stall at temps above 86F , and contams thrive.

Fruiting at cooler temps tends to produce denser, meatier fruits, while fruiting at higher temps will often produce hollow, less dense stems.


Mycelium should be exposed to ambient room light from day of inoculation as has been known for many years.  Light is not a pinning trigger until after full colonization and an increase in air is given, and even then it's a secondary pinning trigger.

The inside of the jar is always a few degrees warmer than the outside because the mycellium produces heat..mycellium tends to stall at temps above 86F , and contams thrive.

Fruiting at cooler temps tends to produce denser, meatier fruits, while fruiting at higher temps will often produce hollow, less dense stems.


Mycelium should be exposed to ambient room light from day of inoculation as has been known for many years.  Light is not a pinning trigger until after full colonization and an increase in air is given, and even then it's a secondary pinning trigger.

appears to have been an illusion. that jar looks perfectly fine to me from those better pics.

You should be more observant -

He posted new pictures of a jar that is fully
colonized.

The jar with the weird green growth is only partial colonized
near the top.

Woops. I thought that was the one in question. This one has some build up from the sterilization process on the outside and bottom part of the jar that I could scrape off with my fingernail. pic 1 , pic 2, pic 3

Massive Update: I was graciously given prints by a member and I have started once again. I did 17 jars and of those,  14 are confirmed as growing and the other 3 with some small pockets (it's harder to see since they're smaller and these particular jars have build up on the outside from the sterilization process). I have them sitting with the lids off in an incubator since it's starting to get cold. I am so happy I made the choice to reach out to you guys when I was worried. Thank you for giving me suggestions, putting my heart at ease, and even giving me a chance on redoing it and getting it right this time.
