How to cap my erlenmeyer flask when cooking agar?
    #19045765 - 10/28/13 11:59 AM (10 years, 3 months ago)

I usually cook my agar in mason jars with filter patches. I just bought a nice Erlenmeyer flask that came with a rubber cork. What is the best way to cap this flask when cooking agar in my steroclave? Should I stuff cotton in the top of the flask and cover with tinfoil? Should I use the rubber cork somehow? Maybe wrap the cork in a coffee filter and then plug the flask? I just don't know the best way to do it.

Drill a hole in it and stuff it with polyfill?

Thanks Anne!

Are you saying drill a small hole in the cork, and fill it with cotton prior to cooking?

So I drilled a hole in my cork, filled it with polyfil and

then tried to blow some air through and I couldn't so i took some polyfil out loosened it up a bit and then was able to

blow air through. I'm now testing this erlenmeyer flask and this new steroclave. I didn't want to waste agar so I put

distilled water in the flask in the steroclave and tested that. That was unsuccessful. The water must have boiled out

the hole in the cork in to the bottom of the pot in the steroclave, then the water got in the vent tube, and no

pressure. If I was using agar instead of water would this of happened?

Nope. I put the cap on tight and pc the agar. Works great. I got it from a user on here Amanita Virosa. He has a mushroom cultivation supply shop in Asheville NC. He has a website with an online store. Awesome guy and very helpful. Google Asheville fungi and check him out

Agar videos on youtube show real labs only using tin foil before putting in autoclave... But maybe autoclave is different... I think they also put an elastic around it, just search for agar autoclave on youtube.

this one is for me since I have no PC
just foil

But I may stand corrected because an autoclave is not a PC so....

I asked the guy when I bought it if I needed to drill the lid and put a filter to relieve pressure and he said no. He said that it was designed just for sterilizing

I've used it twice now and it has worked great both times

We would always put a wad of paper towel  in the mouth and cover it with tin foil.

I just take a strip of foil and tear it in to three pieces and then wrap then over the top.  Works quite well from what Ive been ableto ddetermine thisfar in several batches

Those seem nice. I just ordered 2 of these. Time to step up from the Snapple bottles.
