Petri Dishes to Liquid Syringe to Substrate bag?

Is there anyway to transfer of agar/mycelia from petri dishes to a substrate bag? I don't want to open the bag through the opening because of the tyvek sleeve & don't want to introduce contams. Should I cut a piece out and cultivate a LQ then transfer to bag? or Should I remove the tyvek sleeve after PCing it and just droping a piece in there?
another question is, Can I remove the tyvek selve from the bag after PCing it?
This is confusing at times.

yeah, you should remove the sleeve and impuls seal it or close it some other way. with this you already open the bag a bit, so dropping in an agar wedge wouldn't pose that much more risk, but you might consider the no-pour agar-->lc tek from i think oeric, i don't quite remembter the name of the topic.

Remove the sleeve when you inoculate. Do this in front of a flowhood for best success rate or in a glovebox or still air box as a second choice.
You would be best to drop the agar wedge right in. This assumes it's a grain bag, not substrate. RR

I've injected wedges directly through the side of the bag with 100% success (6 bags). in your sab or flow cab pop the plunger out of the syringe, cut a wedge and put in the barrel, replace the plunger, put on a needle. In open air wipe the bag with alcohol, inject the wedge, and tape the hole with packing tape or autoclave tape.

I pour some distilled water into the agar plate and suck it , then I inject into a LC medium (sealed bottle) and from that I inoculate. I use 1cc for a pint, 4-5 cc for a litter, 10-20 cc for a gallon, I know my proportions are funny. But usually works. Sometimes I use a a wedge of agar and it works. I never used a flowhood but always a glovebox. I have mixed results with grain to grain transfers, Im using Unicorn bags but I fear to leave lots of air in it because air is not totally sterile. I wonder about Rogerrabbit advice of using plenty of air in the bag in the flood hood but how to solve this issue in the glovebox? How much air in the top of the bag and how to avoid contams? Now Im transferring Pleurotus pulmonarius and is fast but my mycelium is 4 months old so maybe is weakened. Any ideas about the air in the bags? Cheers

I just recently did a bag with 60CC of LC and it colonized very fast.

All I did was put some duct tape over the front of the bag to re-enforce the hole and then covered up with duct tape and let it colonize. I realize this isn't best practice but it worked just fine and hasn't caused me any contams.
If you don't have a flowhood I think LC to bag is the safest route, although a decent sized glove box like RR said I'm sure that will work. I just hated doing G2G's or Agar to G in my glove box, it was so cramped.

A good reason to spawn to a second generation of spawn is that it is easy to resolve contamination issues because sometimes the first generation will not exhibit signs until it is spawned to new substrate. This saves spawning bad grain bags or jars to your end substrate that will result in failed crops.

My advice - agar plate to spawn jar/bag. Some people love liquid innoculants but all contaminants permeate liquid substrates quicker than any other substance.
