Accidentally sucked air into syringe
    #19014145 - 10/22/13 03:11 PM (10 years, 3 months ago)

Title says it all. A MS syringe sucked 1-2 mL worth of air. I may just throw it out but I'm thinking about salvaging and innoc a jar of WBS to see if it contams. Any thoughts opinions or experience in this type of accident.

Agar is your best bet.

If you don't do agar I guess you could noc up a grain jar and see what happens.

I still haven't worked with agar. I recently (finally) got a PC and I am finally branching out away from BRF jars. I'm one step at a time and agar is next up.

I don't really understand your situation but I suck air into my MS syringes every time right before I use them for inoculation to mix the spores around better or to break up a chunk if their all bunched together and I have never had a contam until I open the jars or start fruiting and I don't get them very often so your good if it's in that context.

@ spaceman 101.... Do you suck up straight up air? Or like the needle in an open flame trick?

Flame sterilize the needle red hot, then, as its red hot, make sure the entire open end of the needle is inside the flame, and suck in a bit of air while in the flame. Never suck in normal air thats literally inviting contams for dinner, and a couple cc's? Damn.

Ive never done the suck air through the flame trick, but ive read about it.

If i were you i would just inoc some jars as usual, see how they go, if you get some fully colonized without contams and fruited, take prints, and start agar. Inoc a drop onto agar with this syringe you just goofd on  because if there are any contams you can isolate away from them.

I know how to sterilize the needle and all. This was pure accidental isolated incident on my part. I just knocked up 3 WBS jars with the syringe in question, wish me luck on those. As for agar, I put the syringe in the refrigerator to wait for agar to show up. I will order some next week. I can't wait to play in the agar.

well what I do is when I get a new syringe it has a cap over it before you put the needle on so what I do is loosen the cap and suck the air around 1 or 2 cc's before I take the cap off and like I've said no problems at all.

That's good to hear. Hearing that you do that and has always been contam free makes me feel like that syringe of mine has a chance. Thanks everyone!
