Im on my first WBS run as well. The new.location im in doesnt have rye berries, ( A grain store that doesn't have rye! Imagine that.) Im 7 days in and already had 1 contam. Good growth on the others tho. I only used 1 cc per jar and left the corn. I did remove the sunflower seeds. And any other floaters. Your jars look drier than mine and i used the same tek and dried for 2 hrs as well. Good vibes for you.
If you could link to that rye for a dollar a lb it would be appreciated. I searched but cant find it.

I PC'd for 90 minutes after pressure was built up in the PC @ 15psi.
I will definitely keep everyone up-to-date as this goes forward. The MS syringe was my easiest option ATM since I don't have any tissue or LC to use.
About how long should it take to see signs of growth approx? (I know there are thousands of factors that play into this)
Honestly I have had spotty luck with MS syringe to Grains in the past. I do not know were you got your MS syringes or, if they are homemade. Agar that MS, and then transfer to grain. This has been working well for me for a few months.

The OP isn't suggesting that he's having problems yet, his question was about the water in his jars. By all means your idea is a good one, and I always use agar cultures for my edible grows, but thousands of people on this site each year have success with spore syringes, don't they? I don't know, I have never, nor ever plan to, use one.

personal update: 5 days in and no growth yet ): there is NOTHING going on in those jars lol
i went ahead and PC'd 2 more jars tho and im about to noc em up. gonna use 1cc as opposed to 2, and i have 2 noc points this time just to see if that makes any diff :p
edit: ive noticed that my jars have a little water at the bottom, so i must have left the WBS too wet ): if im not mistaken, this is going to slow colonization, and possibly introduce bacterial contams.. so lets hope my sterilization technique was spot on!

The times I have gotten bacteria contams I think are from too wet of wbs when put into jars. Same with when I would mix them with coir and verm in tubs and it would contam before fully colonizing. I think my coir/verm was a bit too moist, needed to be drained more. I personally now lean towards the drier than wetter side just to be safe.

Grains being too wet won't cause bacteria in and of itself. Chances are that if you got a bacterial contam it was either introduced during inoculation (from a contam in the syringe or poor technique) or is the result of improper sterilization. Wet spawn will colonize slower giving you a smaller window for colonization before any endospores that may have survived the PC cycle germinate and take over, which is why proper hydration can help the mycelium get ahead of any endospores. The solution is to up your PC times to kill more endospores and/or switch to an inoculate that is cleaner.

I agree that wet grain =/ bacteria, that is just how it worked out for me. Thanks for clearing that up.
I have modified my technique in my SAB for g2g and that may have been a factor. But I was doing 90-120min at 15 psi each time I pc'd so that should not have been the problem.

Why would you do that when G2G has already been mentioned? Spore syringes are the most inferior inoculation method, they are slow to colonise and a waste of money.
Seriously dude, do grain to grain transfers for three generations and you'll have enough mushrooms to last you and your buddies for years.

just ordered more myself lol. we seem to be on a similar schedule :p
i bought them so i can run a few different strains. i hope to g2g most of my current jars and set a few into a mini mono. i have 1 cc left of both Z+ and GT that are going into an agar wedge in the next few days!
edit: to barong above me. he probably did it so he can noc up the jars he has left, instead of having to wait for his other jars to colonize.. as you stated, MS takes awhile, and if thats the only option atm, might as well make the most of it and pump out a bunch of jars!

That was my exact thought in ordering more syringes. I wanted to use up the free jars and increase my odds of success as well as acquire more strains to pass around.

Personally I only buy a variety or species once. One successful grow and I take prints and clone. The prints go in my collection and the clones I like go in a slant. Then I can either grow that clone forever or, go back to the print to either isolate on agar or do another ms grow and take more clones. Never buy anything more than once. Even a ms culture plate on agar is good for a few months. I never go with a syringe straight to grain, have had too many dirty syringes.
I'm at the point where I got enough varieties and species to keep me busy for a long long time Truth be told, most of my collection came from the marketplace anyway This is the best way to be self sustaining IMO.

Chili, Why do you want more 'strains'? They all do the same thing.
Anyway, I'm just saying, once you try G2G and see how fast it takes off (therefore increasing your chance of success) you will never by spores again. It's simply not necessary. I'll posts a pic of a 6lb bag I colonized with gen3 spawn later. Four days on, its alreay half done

UPDATE: two days later and every jar has growth in it. Two jars I am questioning if a contam is present. But the rest look great.
I can't get a great picture because it isn't heavy yet on all of them. But here is one..

I know its a long way from success, but this is a good start.

Everything you see over the next week or two, you are going to ask 'is it contam'? Generally, you will know, because it won't be white. Then you'll discover information about a white contam known as 'cobweb' and start asking the question 'is this cobweb?'.
Be patient during colonisation. You'll know if all is well. SO far it looks to be the case, so well done

Thanks. Yes, I have read up on cobweb. I read that it spreads very quickly and you'll know when you have it because the entire jar could colonize within a couple days. That would obviously not be mycelium because it takes much longer for it to colonize.
Will keep posting updates.. Two jars definitely have trich. Took em out. Threw em away.
I dont care about saving glass.. It's not that much anyway. Tossed the entire thing. Screw it.

Happy you got growth after the ms noc and shake! When i started reading this post i was concermed that you may not have let the jars cool enough enough after the pc, only 2 hours. I did that with some of my jars a few weeks ago and 1 in three did not take, even with no shake. Now i just let things cool in the pc overnignt, and have not had a problem again.
Good luck with the grow and let us know when you shake, how long it takes to recover (for me two days usually) and once its finally @ 100%!
