Taking mycelium from a colonized grain jar?

So I think I may have heard someone mention something before about taking mycelium from a colonized jar and using it to make a viable syringe or use to inoculate a liquid culture. I currently realized I had used the rest of my spores but I have a fair amount of colonized grain jars. I want to know if anyone has successfully pulled live mycelium from a jar using either peroxide or sterilized water, injecting it into the colonized jar, swirl it around, and then proceed to draw it back out the jar via sterile syringe? I would like to know if anyone has done anything similar or if anyone has a link to a tek. I had no search results, or perhaps worded it poorly. thanks and much appreciation

The only good way to use grains from a grain jar would be to open them up in a SAB/GB/flow hood and put a grain onto agar. You could make more grain jars and g2g a colonized jar out to keep it going as well.
IE I wouldn't bother doing anything you said. Maybe you get lucky and it works

I did just do some grain to grain transfers yesterday since I am out of spores at the moment. I did it in front of a very large laminar flow hood as well. The only thing I am afraid of is the afterthought that I did not use peroxide. Do you think this would be an issue in G2G, even under extremely sterile conditions. Its only been 24 hrs and I already can see the mycelium reaching off the original grains onto the new ones.

What would you ever need peroxide for in this hobby except in the rare case of cobweb on a substrate in fruiting conditions.

How do you even use peroxide for a G2G anyway? You have a laminar and are doing g2g yet you cant find a way to save mycellium from a grain jar?

well more or less since I dont do agar and I dont always have access to the laminar flow hood, I need a way to store mycelium until I can generate a spore print. So I want to see if there was another possible way. as for the peroxide, it is used as an agent to help prevent contamination when doing g2g transfers. According to Nicholas and Ogame; "Grain-to-grain transfers are performed more or less like agar transfers: After adding peroxide to the sterile grain jars, a small amount of colonized grain is poured into them." Now the book does not state whether it will affect the transfer without the peroxide so I was just curious as I have never done g2g transfers until yesterday. So again I am curious to know if pulling live mycelium will work

Well shit you need to take a look at this
What you need to know about g2g
Never heard of h2o2 being used for g2g seems like just another route to get contamination.

Yes you can do this. The results are very iffy. When you try and soak/suck mycelium into a syringe you might also add contam to the jar and the syringe.
Process/ You need a clean syringe and Jar to make your LC in.
I would boil some water and clean out my syringe. I would also make a honey water mixture and PC it for 30 mins at 15 PSI.
Next you take your Clean syringe and flame sterilize the needle, then poke it through a jug of spring water. I like to rub the side of the jug with an alcohol pad before sticking my needle in it. I fill up my syringe with fresh spring water.
Then in the SAB, flame your needle, and open your grain jar, spurt and then suck up your mycelium. Add it to the jar with the Honey/ Water that was PC'd.
Wait 5 days and you should have mycelium in your LC.
The problems
With a LC, it is very hard to tell if you have contaminates. Most people will advise you to transfer your LC to a Agar Plate. This will allow you to make a few transfers to reduce/ eliminate contamination's.
Check out RR's Vid's. He has a great section about Agar, and the benefits of Slants to keep your mycelium for up to 2 years in a fridge.

Ahhh! nice man, I like the LC tek idea. definitely gonna try it. I do have a 24x24x12" HEPPA filter and a 760 cfm squirrel cage fan, I just need to get some nice wood and dedicate a few hours to building the flow hood. I gotta admit that I do enjoy trying new ways and methods of obtaining fruits and storing genetics/spores and I have learned a lot about what works and what does not.. but its always nice to learn from those that have more experience in the field. BTW is agar difficult or relatively easy once a flow hood hood is built?
