what am I doing wrong

Hi all I desperately need some advice or ideas. My first attempt with 1/2 pints jars verm and brf went well. Used syringe and successfully started good colonies with some nice aggressive mycelium (Cambodian and GT strains) got fruiting bodies and made new syringes. My problem is on my 2nd, 3rd and 4th attempts using same methods, I have only managed to germinate some weak spores and any mycelial spread seems to weaken and stop. My latest attempt(4th) I made sure a only very lightly packed the jars(as I suspected poor gas exchange and too tightly packed medium)and used two new syringes I ordered recently, one Cambodian and one Giant white. I also inoculated two separate jars with last years home made Cambodian syringes( one whole syringe per 1pt jar). Its now about ten days later and both 1pt jars have germinated ok colony the rest half pt jars show no sign of life. The jars are in a dark container that's set in water heated by ft heater to around 28c. Wondering why I had such good colonization and healthy robust mycelium first time, then poor slow growth if at all on subsequent attempts. Cant say Ive significantly changed any of the process, no sign of any other infections due to poor sterilization. Any ideas

how long after you sterilize your jars are you waiting until you knock them up? also are u following your tek properly? could be that you dont have enough moisture in the jars to colonize.
p.s. - you shouldnt need to incubate your jars, they will colonize just fine at room temp with ambient light

Take your jars out of the dark heated container. They will do fine on a room temp shelf with ambient light.

They colonize faster with higher temps and a lack of light doesn't hurt them when colonizing. Just because you don't need an incubator doesn't mean it can't benefit or you should not use one if it's already on hand.

yes last year I had success with the same technique. I had fruit so I made spore prints in sterile baggies then made syringes from those. I seem to be having some problem with the substrate. Even when I get germination the mycelium seems weak. Last year I even transferred mycelium between jars, from a jar that had robust and thick mycelium to ones that had nothing. The transferred mycelium quickly took off and colonized the new jars. This time (2nd 3rd 4th attempts) , nada. It really seems like the substrate is different somehow. Thanks for any ideas!

Thanks! Im pretty sure im doing things right. No contam but nothing else either. Jars cooled 24 hrs seem to have nice moisture, no excess in bottom of jar. Used 2 new syringes and two home made. Home made ones germinated in two days , but the new store bought ones, there's nothing.

Ive never seen spores germinate in two days. Those home made ones might be funky if you saw growth that fast. Keep an eye on them.

I have seen spores germinate in 2 days, but scared me I agree keep an eye.

Ok, so let me get this straight,
1st grow went ok
2nd, 3rd showed only slight myc growth at the point of inoc., and then stops
4th grow, you used 1 syringe per pint jar, and the growth seems good
Is that what I'm reading?
Did you remember to add water to your BRF/ verm? If your jars were very dry to begin with, perhaps the syringe is hydrating your substrate only at the point of inocculation.
This is only an educated guess. Pics might help.

Ya 2 days might be a little exaggerated, maybe three or four at most but defiantly is mycelium. But my problem seems to be why im getting such different results after doing same technique. Are spore syringes that notoriously unreliable? Jars are free of contams cooled at least 12 hours, inoculated without issues, but no germination and no growth if mycelium is introduced. Thanks for your ideas and thoughts !

almost right, this time my 4 try, I used 12 jars consisting of 2 pt jars and 10 1/2 pt. Substrate was well watered before and after sterilization. Inoculated 5 jars with one Cambodian and 5 jars with Giant White. The two pt jars I inoculated with 1 homemade syringe each. Put the full syringe in the jar. Both these showed germination right away after 2-3 days. The other ten nothing. They are in incubator

2. How how is your incubator if it's like 78-80 in there the jars are too hot as the mycellium makes heat too.

Aha, well, it was just a guess.
So, the ones that were shot up with homemade syringes are the only ones showing growth?

in one post you say 24 hours in the next you say at least 12 hours... which is it?

i know i'm gonna get shit for this but try taking them outta the incubator, and just put them on a shelf with ambient light. incubators work great, in fact i'm thinking about building a new one once i get my isolate going..., but if you're having problems with stalled growth lets start eliminating some variables; such as you are incubating at too high temperature.
