Bacteria on Agar

Hey guys I've been trying to isolate a good section of Mycelium and I keep having issues with small amounts of bacteria coming along for the ride to the next dish.

RR suggested in another thread "You can also pour hot agar right over the bacteria on the dish. The hot agar will pasteurize the bacteria, while not seriously harming the mushroom mycelium, which will grow up through the agar to appear on top, generally free of bacteria. I use this trick a lot on outdoor mushrooms I bring in to clone."

So pouring hot agar onto the mycelium isn't going to kill it? I was under the impression that hot temperatures would kill the mycelium.

I was having serious bacterial problems with agar but then I started dipping the piece in peroxide for about 3-5 seconds right before putting onto agar, I do this all in an SAB

No bacteria since

What % Peroxide were you using? I'm doing all my work in a SAB and I'm fairly positive the bacteria is being brought along with the mycelium sections into the new dishes

I just use it straight out of the bottle, I think it is 3%

When you say dipping the piece what piece are you referring to?

You could test your technique first by doing some dummy transfers. If that's alright then you should be able to get a clean culture by taking a tiny piece that's furthest from any contamination. It can take quite a few transfers.
