Sterilization port in SAB

So I've recently revamped my sterile procedure because I was incredibly naive in thinking that my old SAB was worth a shit. After many contams that I was contributing to the new environment I'm in that has dogs and such I really took a hard look at what I was doing and built a proper SAB.

Now I'd like to continue my agar work, poured my petris yesterday and made up some spore syringes too. I managed to get an isolate from my previous runs with the old SAB, but it is dirty. So using the new SAB and my fresh Petri's I plan to get this thing cleaned up and in proper action.

I'm running some trimmed tyvek sleeves into the SAB. I'd really like to avoid pulling those in and out to flame my scalpel. I'm looking into picking up a bacti-cinerator but that's down the road and I'm still not sure I like the idea of it inside the SAB. I'd have to check the flash point of ISO and the other chems to be comfortable with that.

So the idea is to drill a small hole in the back of the SAB that's at an appropriate height to put a scalpel through and sterilize by torch. The torch would be sitting on the outside, perpendicular to the hole. The hole would be covered on the inside with a small vinyl flap so it's not permanently open. Just lift, sterilize, pull in and drop the flap. Thinking a 1/4 - 1/2" hole.

I have seen some talk about this in other threads but not a lot. Most people prefer just pulling in through the sleeves. I would like to avoid bringing in a red hot scalpel through my scrunchy tyvek sleaving.

The issue I see that could happen would be airflow out of the hole and igniting via the burner. I'm thinking I'd place the burning 2-3" away and have to really stretch to get the tools to it. And with little to no airflow, I don't see that catching the SAB on fire.

But, what do you all think??

You're over-thinking it...

Sleeves should not be attached to the still air box. Doing this creates a pressure differential when putting your arms through which can bring in contamination. Use the sleeves on your arms if you must. Without the sleeves on your still air box, you can easily flame your needle or scalpel and get it in while still hot and sterile.

I use my still air box in a very unsterile environment with no contaminations so far. I would re-examine your procedure before changing your equipment.

Your still air box should have holes that are generously larger than your arms, most recommend 4", so that you can place your arms in/out with minimal air displacement. The holes should be placed on one side, and there should not be any other modifications. Simple designs usually work the best.

I built mine Ala' TL's SAB tek.

I don't know if sliding gloved arms down attached sleeves is any different then sliding sleeved arms into a port as far as air is concerned.

But, I can take off one of the sleeves I suppose.

If you built the TL style box you can leave the sleeves attached no problem.  I always found it easier to leave both sleeves attached when I wouldn't be moving in and out of the box (i.e., no flame sterilizing such as when pouring dishes) but if I were going to be moving in and out of the box to wear one sleeve on my right arm and leave the left sleeve attached to the PVC like normal so I can move my right arm in and out without creating pressure changes.  In that scenario my left arm would always stay inside until finished.  Obviously swap those if you are left handed.
