Could this be wet spot..?

Hi all,

Below is a picture of my wheat grain jar, spawned from fully colonized Ecuador grains on the 5th of October. Does the wet spots resemble wet spot? i'd hate to lose a jar so nearly fully colonized... The Grains were soaked +-24 hours, with gypsum, and then slowly boiled for 30 minutes, an then jarred. The lids were micropore'd twice over where the 4 GE holes are. Then the jars were autoclaved at 122 degrees Celsius for 2 hours. Pressure was about 1.3 MPa. After cooling down the jars were inoculated via grain to grain in front of a HEPA laminar flow hood. I shaked it on day 3 as it was fusing nicely with the surrounding grains. Thereafter we had unexpected shitty weather, so I popped them in my closet with a thermometer and a 60W incandescent light bulb, the temperatures usually hanging in the range of 26 to 28 degrees Celsius.

Working with grains can be tricky because you have to rinse and drain them to the point that they are not slimy or overly wet yet you can't let them get too dry or else the myc won't be able to colonize as fast. I should know, I've screwed up in both directions :wink:

In my experience a little bit of excess moisture isn't so bad for a grain jar, provided you have sterilized it in a pc/autoclave, inoculated it with clean materials, and the seal on the jar is sound. I have had fully colonized cakes that had a little bit of excess water in the jar that would squeeze itself against the wall, and these were birthed with nothing wrong whatsoever.

Wet spots are more of a casing issue. Besides, there isn't really any "maintenance" you can perform on a jar other than tossing it. I would say keep an eye on your jar, but do not dispose of it unless contams become visible. And trust me, when you have actual contams, there is no second-guessing.

The grains should look completely dry when you load them in the jars. You'll know if they're too dry only by the size of the grains which is related how much water they've absorbed. A little extra moisture will have a far larger impact on colonization than a being a little too dry.

OP, that jar looks like it was probably made too wet but there's also bacteria at work there, there's why the wet spots are getting a slimly and bubbly appearance.

Here is a pic of the jars again,now a day or two later. Is there any way of saving them? I wouldn't want to lose it. What if I added some H202 to try and kill off the wet spot? I suppose it is from having to many burst wheat kernels? I am preparing a new batch now, attentively making sure it doesn't boil too wildly and burst kernels again. They have been soaked in gypsum-water for about 28 hours beforehand.



Below is another pic, taken before the weekend, before I threw them it the closet with the light bulb for heat, and also before the appearance of the wet spot.

The bacteria could have been from improper sterilizing or making the grains too wet. Most likely a mix of both.

There's really nothing you can do about it at this point besides PC them again and dump it outside to reuse the jar, or make a small outdoor bed with a bit of substrate and hope you'll get some fruit.

Peroxide has no place in dealing with contams other then Cobweb mold.

I doubt it if you inoculated the jars only 11 days ago. There may have been some bacteria in the master jar you used or it got in during the transfer. It's a good idea though to try not to let incubation temperatures rise above room temperature and limit the soak time for your grains to no longer than 24 hours.

Yeah I won't make that mistake again. Planning on going TiT, just got myself a nice digital thermostat online!:rockon:

And I am also sure the master must've had some dormant endospores waiting for fresh grains...:sad:

Thanks for the advice Jimmyhunter1000. I did a quick smell test, and they both smell like good ol' earthy mycelium, so I will let them ride it out, pasteurize some bulk substrate on the drier side of field capacity, at a 2:1 ratio, and see how it they fair!
