Does the ideal LC Recipe ever change?

Trying to get into growing edibles and just knocked up some ryeberry spawn bags with stropharia. Only a little LC left in the purchased syringe. Can I use similar LC recipes that are used for cubensis (Sugar in the raw is what I want to try because it's available to me) with the rest of the edibles? I don't have a flow hood and think using the last of my LC to start another LC would help me stretch out this species I purchased so that I can grow and return to this species over and over. Does anyone have advice on building a 'gene bank' of sorts? I know if I am going to grow a few strains I do not want to purchase the same 3 LC syringes offline over and over.

you can try...my luck has been minimal. i would just suggest using proper injection and breather ports over large filter disks. but i've been really good at growing yeast with LC...

do u mean for stropharia in general or other gourmet species in LC? Why would it not work if you already have mycelium in liquid.

Because liquid culture easy to contaminate.
I won't use lc anymore anyway. Agar is much easier and faster.
If you drop a wedge of agar in a jar of grain, you get growth so fast. LC has always taken forever to colonize a jar of grain. When I order LC syringe, first thing I do is knock up a few petri dishes. Then use those to knock up grain.
Edit: I don't have a flow hood either, but haven't had a contamination yet!

I have plenty of agar... but my SAB is very primitive (a tote with 2 holes cut in it) so most of my petris got contams when I tried. It's such a useless SAB, that I just shot up my cakes in the open air a couple of weeks ago. I just lsyol'd the room, wore gloves+surg mask, and wiped everything with alcohol napkins. Should I order more petris and try again? How is your SAB/Glove box set up? I'm really not confident in my SAB and I suck balls at building things. I need to get better at this though because I'm mainly interested in gourmet mushrooms and the sterile technique to do agar plates and g2g seems a lot higher than just inocculating cakes with MS syringes.

Get more plates, but get glass or polypropylene ones so you can premix agar and pc them. My SAB is literally a 15 gallon unlidded tote on its side. No arm holes, no lysol. I just keep my work in the rear of it, limit exposure to air, never pass my hands over an open plate.
If you need help I can help you figure out agar.

Sounds good. Do any shroomery sponsors sell the polypropylene petris? I was going to buy the plastic ones from mycosupply, but if you can pre-mix and sterilize that sounds really awesome.
I guess I can try regular plastic ones because they come sterile too though? I don't have to open them until I have my agar ready in the SAB. I've done this once before but I think over half of them contaminated, I could probably do it more eloquently this time around as it's been a few years.

None of the sponsors sell autocalvable ones. I bought a bunch of PP culture jars I use for tissue cultures as well. But I autoclave cheap little glass ones too, they cost about a buck each on ebay. When pouring hot agar into a dish in a still air box, a lot of air turbulence is created from the heat rising off of the plates. That's why I premix and then pressure cook my plates.

Can you post a picture of the PP Culture jars? You are saying that you use small glass jars and PP jars and sterilize them with agar in the base so u don't have to pour petri dishes? I bet that works good, unless I misunderstand what you are doing.

I use presterilized petris in a sleeve and pour hot agar into them but I agree with liamtheloser about pre pouring then sterilizing. It does seem to work better when using a still air box.

You can pick up small PP Petris at tedpella.com for really cheap. I got the culture jars from a seminar, they're like 50 cents each.
I use micropore tape to close my Petris and cultures, it much cheaper than parafilm
