I have been having some poor results from poo. I think I may have gotten some that was to fresh. So I got to thinking, maybe there is a better way to do this, increasing yields, and reducing effort.
I mentioned this idea once and people said the rye may not have enough moisture. Let's find out first hand.
??/??/ Agar was colonized and a sector transferred to LC. ??/??/ Lc colonized well and was refrigerated.
10/7 42 grain rye jars inoculated for the experiment.

Well, I've never had any significant results from straight grain casings. coir is easy, why not try it?
and unfortunately i see a good amount of burst grains :/

You could just go with a CVG sub and call it a day . If straight grain produced as well as spawning to bulk, you would find almost every TC on the board doing it.

I cased one quart jar once and got an ounce on the first flush dry so if it does anything comparable to that per jar then it will be a winner.

My reasoning for why I think it will go relatively well, specifically the cased tub, is every time I ever used more bird seed grain spawn I see a significant increase in yields. I once used 9 quart jars of bird seed spawn and 8 quarts of poo and got an 11 ounce first flush vs my average for 4-5 quarts of spawn and 11 quarts of poo which would yield an average 6-8.5 ounces.
Rye is suppose to be superior to bird seed so my end result should be I am hoping 12-3 ounce first flush. That is what I am at least hoping for. It will be less than a ounce per quart but it is what I have my expectation to be.

Is this an isolate? You say you transferred a sector of agar but didn't mention if it was. If it isn't, then your results cannot be conclusive.

It is not an isolate but if it yields well it will show that it is a method to grow.
Everyone is so quick to dismiss even trying it without any evidence to support that it is a bad idea.

Well, since it's still MS there is no real credibility on a single test.
Maybe if you did a hundred or so with the same experiment, using MS, you'd at least have some patterns to try to sort out.
Why not just get yourself an isolate?

The easiest way to get a good-fruiting isolate is to clone a nice cluster from the first flush of one of your MS grows.
Isolate any sectoring and then test them out. IME, 4 out of 5 turn out to be great performers. Saves a lot of hassle when you just let the myc sort out the best fruiters for you.

What if you just take a good cluster clone and not isolate it any further? How will it perform?

Some will be isolates from the start. You can certainly do a straight tissue to agar to grains, and it will perform how it performs.
This issue with a clone that has multiple genetics is that you cannot save that culture for later use, without a lot of unnecessary work trying to keep sectors together.
And since it is not uncommon for your first plate to be dirty, so sometimes you have to make transfers anyhow.

Yeah I took a clone to agar a few weeks ago and ended up with 4 isolates. I took them to grain masters about a week ago.
What would you say the average # of strains per clone is? 4 or 5?

I couldn't tell ya, I never kept track 
I just take a small cut from the leading edge of the fastest, healthiest looking mycelium after it grows out an inch from the original tissue.
