Prepping my rye berries now so I can PC them and let them cool over night. Have a P. Galindoi print I'll be working with and was curious to see what people thought about the whole rehydration of spores concept.

I planned on just making a syringe tomorrow when I threw some of the spores on agar. Would it really be worth my time to go ahead and make the syringe today so the spores have a bit of time to rehydrate themselves or would throwing one together 2morrow then using it afterwards really make much of a significant difference?

I assume if I allowed them a bit of time to rehydrate they would show sign of growth sooner however considering I'm shooting for stones in this particular grow a few extra days will go unnoticed.

What cha think?

They shouldn't need to be rehydrated. Also, if you're using agar, why make a syringe?

Just do spores to agar and go from there. A 3 month old print shouldn't need to be reydratrd for it to terminate. I have a 8 month old print that germinates in 4 days with just the spores to agar

For some reason I've had shit luck with getting straight spores to germinate onto agar. Moving a colonized grain or doing a clone has been no issue at all though. I've just been using a LME and agar mixture using Doc_T's tek:

What particular mixture do you use for your agar? Is there one you've found that work especially well for spore to agar?

I figured Id make a syringe and see how that worked out and throw some onto agar as well to see if it'll grow out for me. I guess I like to over prepare myself... If the agar grows out then I'm more then likely going to make another set of jars to put aside using that and just keep that alive and moving while I work my way down to an iso if possible.

I use mea. But golandi doesn't like it. It still works, bit doesn't grow as fast
