well as some might know i recently decided to try agar again sadly the 3 i attempted from spore syringes never germinated anyways i decided to inoculate another three petri dishes with a piece of colonized grain when i did my G2G. i made 1 AA+ and 2 PE and now here they are.
   the circled areas of the PE plates contain what looks like lipstick mold and i was wondering if the diamond areas would be the prime choice for the first transfer? as for the AA+ i figured just about anywhere would be good but most likely right under the label seems prime. thanks yous guys

Yeah on those first 2 plates just try to get your transfer as far from the contam as you can.

The top right looks more promising. Id take wedges from that and inoculate more plates.

top right of which one? and about how long can i have grain jars sitting before inoculation? ive read forever and ive read a couple weeks is why i ask as well as the fact that i have 15 wbs jars that where pc'd on like 8-26 and 20 rye jars that where pc'd on 9-6. both wbs and rye where prepped via simmering.

It kinda just depends upon how well the jars were prepped, if they were simmered it's unlikely they'd last anywhere close to a year, perhaps not even a few months. The first thing I'd look for is liquid pooling in the bottom of the jars, that's an indication of bacterial growth. You could always try a test on one to check, but then you're also risking contaminating the spore syringe if a contaminant happens to makes its way in there, which isn't particularly likely, especially if you sterilize the needle afterwards, but it's still possible so it's your call. If you want to be absolutely safe just make new jars.

Mycelium performance and appearance is not an indicator of the genetics you want.
Aggressive mycelium on agar does not indicate fruiting or potency potential. You can't tell by looking.
To get the genetics you want, you will need to test all the sectors.
You can take samples fairly close to contamination. You don't have to ditch a sector just because it has a contamination.

this is for the most part just practicing with agar and working on my technique. i plan to take clones from my current grow once i get fruits based on size, clustering, and in the case of the AA+ albinism. these clones will be isolated for future grows. in the case of the jars im more so questioning them drying out. i will be G2G'ing and will take a nice sniff before inoculating aswell

It has been reported many times that isolates made from clones may not perform as well as the original clone.
Theory is that multiple genetics have worked in concert to make a good fruit, but, perform poorly when separated.
Kind of like when Phish broke up.
