Dry spore print to agar. Did it work?

Literally, I've used inoculation loop before and this was arid dry. I gather microscopic collected on its way to the agar but it was a dry futile seeming scraping endeavor leading up to that.

I have done dry print straight to agar many, many times using an inoculation loop.

Here's what I do: After flame sterilizing the loop, immediately cool it down in your agar plate. It will pick up a bit of the liquefied agar solution and the spores will stick much better to it when you rub it on the print than if it were dry.
I have also done it successfully with a completely dry loop and a simple swipe on the print followed a simple, very quick circular motion on the plate.

Keep in mind though, depending on the freshness of the prints among other variables, they make take quite long to germinate. If it is an old/very dry print germination onset will be delayed -- the spores need to rehydrate.

I am sure your loop carried over at least 2 spores up no matter how dry it seemed.:grin:
