Spore death at poor temperature

so, I purchased a few myco bags and then some spore syringes and set them away in a miniature chamber made from an old suitcases after inoculation and provided a steady temperature with a heating pad. After three or four days, and a sense of impatience, i saw no mycellium growth and decided to apply the heating pad to a direct low temperature on the grow bags from beneath, similar to as when used for seed germination. I checked on the grow bags the next day to find that they were piping hot to the touch and practically steaming inside. I read up on a few overlooked details and figured that all the spores were probably cooked out at the point of temperature as I found them and decided to inoculate each one again about an hour ago and to remove the heat source to an indirect application. Now im waiting for better results. Wish me luck.

This time don't use the heating pad at all. They will do just fine in room temperature.

If you insist on incubating, definitely discontinue once your bag begins to colonize as large amounts of colonizing spawn will put off a great deal of heat on its own and you can easily "cook" your white fuzzy friend.

Ill have to remember then to reference that detail when I begin to notice mycellium growth. After already nearly losing one batch, im hoping to reserve the rest of my resources for another grow if this one goes successfully.

You'd still dry out the substrate incubating like that. A heating pad should never be used.

I will remove the heating element immediately then. thanks everyone. Probably the reason why there was no growth to begin with was because of the unnecissary heat. Ill let you know how it goes.

its only been three days since the most recent inoculation on each of the grow bags and upon checking them, ive seen no real progress to no great surprise, however, Im worried about how they may be too dry. now, i have an empty spore syringe that can be utilized for including moisture, however im worried about contamination. The grow bags consist of a wild bird seed and and a mixture substrate. the 2 mixtures seem to have a decent amount of moisture without pooling and without many droplets. The wild bird seed however, seems very dry although i notice a few droplets. Is this normal or should i perhaps rehydrate each container? thanks.

I wouldn't add anything to them. You could crumble the grains up in the bag to redistribute some of the moisture that is in there.

Theres been absolutely no progress with the myc bags. Im guessing its either due to the myc bags being of poor quality to begin with, or if the spore syringes are bunk. I prepared, sterilized and inoculated a jar yesterday and so Ill find out after a few days if it was the spores or the substrate.

Did you inoculate against the side of the bag, or into the center of the grains?
If it was the latter, it is possible that you have growth, but it hasn't reached a spot where you can see it yet.
Just be patient.

I followed common procedure and just inoculated the substrates again with a new spore about an hour ago so i dont expect to see any growth for a few days.

You might not have even needed to inoculate again. I've had growth take up to three weeks to show up and I've heard of it taking as long as four weeks for some people.

For exactly the same reason, I left one substrate container exactly as it was to determine whether or not the original spore samples were simply slow to colonize.

For sure, even if that one you left alone doesn't colonize though, that doesn't mean the other ones wouldn't have. There's no guarantee that every single one will colonize. It's luck of the draw with MS. Hope all goes well though.

Theres been absolutely no growth from any of the substrates. I even prepared a pf jar (with correct proportions and sterilization) with absolutely no result (not even a contaminant). I have about six syringes and a mono tubs worth of useless substrate. Im horrendously discouraged, however, i must thank the forum for the amount of guidance given, despite the absolutely fruitless results. Heck, Perhaps I may have better results with botany.
