Making grain masters from agar wegde

Do you guys think it would be advantageous to use pint jars instead of quart jars to start new grain masters from agar wedges?
I was just thinking using smaller jars with less grain would result in faster colonization times. It's not that I am impatient I have just noticed it takes forever for a qt grain master to colonize from an agar wedge. The way I see it faster colonization is always better. What are your thoughts on this?

Just shake your quart jar when at about 20-30% then growth will explode. You will have very little room in a pint jar for a shake and not much grain if you leave room at the top.

Agar to grains is always a bit slower than I would prefer, but once you shake it it will take off.
I keep my jars incubated at 80-84F and they are done in two weeks.
You can also slice your wedge into multiple wedges, though it increases the risk of contam each time you open the lid.

Not a bit. I think you would be surprised at how fast an agar transfer colonizes. On the 11th of this month I did my first agar xfer. It was an ms agar grow out to verify "good". The jars were at 100% last weekend. Hands down the fastest I have ever had that many jars colonize so fast in unison. Every one of them was colonized at the same speed.

I have done agar to grain so I know how long it takes. I guess g2g will spoil you with how fast it is

Maybe I need to build me one of you tit incubators. My colonization temps are about 68 or 70.

I wouldn't go over 80F in the room, especially if you have a lot of jars. Remember, glass is an insulator so it's warmer in the jar than outside. RR

When I have my tit incubator set up, its only for petris. Otherwise, I have a lot of bacteria.
I would suggest a qrt jar master grain, then G2G into 10 qrt jars, G2G 9 of them into spawn bags, and refridge the extra until needed
Or just make sure you have a consistent cycle going so you have myc at every stage to keep a consistent harvest

Oh help me Rhonda, I am tripping tonight!
Oh yeah I just threw some agar into a master jar. Slow Slow Slow is the way I shook her.

My TiT sits nicely at 80F.
It was at 86F for a month and a half (oops), during that time I had zero contam issues
I wouldn't recommend going that high though. 80F is perfect IMO.
I don't think heat causes contams. Contams cause contams. Heat can damage your mycelium though.

It never gets cold here and I try to find the coolest place in the house for colonizing grain jars.
If my jars get over 80 F, any bacteria present really comes to life.

If any bacteria is present in your jars, they are contam'd at any temp
Good luck psilly!

The fact is, running an appropriate temperature for the species is very important. If we're growing E coli, we'd use a warmer temperature near our own body temp in order to favor the organism we want to grow. If we wanted to grow thermophilic bacteria, we'd run around 90F to 100F.
Most mushroom mycelium will grow fastest at around 80F. However, that can also stimulate competitor organisms. It's not just about sterilization because the material we use for filters is rarely 100% effective, especially those crappy postage stamp filters on spawn bags. We need to assume that contaminants will get into our media at some point and adjust temperature and other parameters to compensate.
For example, my Shiitake blocks spend three to four months in the bag before stripping the bags off to fruit. To compensate, I keep my colonization area at 60F to 65F, which means slightly slower colonization times, but substantially lower contamination rates during that three to four months.
For cubes, a temperature of 75F to 80F gives the best performance since even on a petri dish there is no increase in lateral colonization speed above 81F, and it begins to decline in speed at 83F. Temps above this amount stimulate molds while throttling back the cubensis. RR
