Question About My LC Grain Jars
#18899057 - 09/27/13 02:54 PM (10 years, 4 months ago) |

For the past 2 evenings I PC'd 7 jars/night and I have 2 LCs I made from LME only that look pretty good and so I inoculated each batch with a sample from the two different LCs..
The jars are showing puffs of mycelia and all jars except 3 were inoculated with 2cc's, 1 with 4cc's & 2 with 5cc's (experiment)
The mycelia would be great news of course but the pooling water is looking really nasty at the bottom of some of the jars.. i don't have the resources to take good pictures right now but i'll try to get them on here.. it may be mycelium on a sunflower seed but it's got a grayish appearance to it that I do not like.. it's not in all the jars either. I'm thinking I should separate them so please respond whoever as soon as possible & help! thanks -Pk

If there's no pictures.. and judging by your tone I can only say that yea they probably are not good if your instinct tells you they're not.

That "clean" jar looks awfully wet. Could be your grains were too wet or you used too much inoculant. Looks like bacterial contamination. If they keep looking a bit slimy, or they develop a strange smell, you can bet that's what happened.
I will never use LCs until I have my sterile technique down 100%, and even then only from perfectly clean cultures on agar. Contamination risk just too high imo.

Retaardvark - Thank you, yes You're right it's wetter than wet.. I'm positive that's why these jars contamed.. I haven't been doing this that long but I've made enough grain jars to know pooling water causes bacterial nastiness such as seen.. I know that smell too but I'm not opening em lol I just PC'd all the visibly gross ones for 90min @ 15psi. Also I'm pretty sure ,in some I used too much inoculant.. the picture with the LC jellyfish at the top of the culture is a GLC > LC (LME) which I didn't use yet but my God look at it compared the the others! I'm gonna try that but as a rule I'm not liking LC, I did open air G2G (just to use a very waterlogged grain jar I made the GLC from)& it was so much less of a hassle & quicker.. I try to exercise extreme caution in my sterile procedure but no ones perfect. Lot of variables this time too using LC and different additives (gypsum, liquid coffee & hydrated lime in soak water).
Screw it better luck tonight.. best soak those grains

I never did quite understand why excess water causes bacteria. I guess the only answer since we use sterile water is our grain jars never get 100% sterile and still harbor endospores.

It's because the mushroom mycelium can't colonize it. Any uncolonized food is going to get taken by some organism. Sterilization is never complete, but simply a window to get the grains or whatever colonized. RR

Maddchef - I've wondered the same I figure bacteria thrive in those conditions vs field capacity being the conditions for mycelia to thrive
RR - Makes pefect sense, thank you for chiming in.. There was still grayish black residue at the bottom of the jar after PC'ing again for 90 minutes (cooled o/n), dumping the grains, filling jars w/ hot h20 & soap and vigorously shaking/rinsing them.. they're in the dishwasher now & then I'll PC them again just to have peace of mind
Leftover residue harmful? Yea or Nay? Thanks again guys

I love my lysol man! That & Iso has kept my work clean for the past 4 months or so and my house is not ideal by any stretch..
I just made my own lids w/ SHIPs too.. now I'm leery about using them again..

Hey RR can you tell me if after all I did to contain/clean this contamination upon discovering it, if it's alright to use those same jars & lid w/ the rtv & sfd i made? there are black spots still at the bottom of the jars even after a 90min pc & dishwasher w/ heated dry.. is it inert material or could the bacteria still be kickin?
