Problems on agar...UPDATE.. I have a theory.

Hey guys...having an issue here.. I did transfers of pan cyan mycelium and also cube mycelium..the pan is on the left. I did these transfers a few days ago..as you can see the cube has colonized the wedge that I transferred but has not climbed off onto the new Agar... and the pan mycelium hasn't moved AT ALL since transfer....Is it possible I over cooked my agar? I've read that I would have an un pourable mess if that was the case..is it possible to reheat the same agar too many times?

seriously...what's happening here.

With cubes and pans you should see movement on to the new agar within 48 hours.
Nakor, it seems like your agar is not hospitable to your mycelium for some reason. Did you do anything different with these dishes? What media did you use? Did you swirl the agar in the bottle every so often while pouring?
You can just grab those wedges off the plate and move them to new ones.

I would imagine it's the same agar he used for the original dishes that it did colonize on since he was asking about re-using the same agar.

Your agar mix is probably too much nutritious. That is one possible explanation but you need to give us some more details: agar mix recipe? Temperature?

I use the pre mixed MEA anti-biotic agar powder. I was pouring plates nd my agar got thick while I was pouring this plate... notice it's lumpy, I used it anyway, idc,lol.. so this was done the same day from the same bottle...the difference is the two in my op where done with agar that had re-heated again after that lumpy plate was poured.. as you can see the lumpy plate is just fine..

Weird Did you let the agar cool after pouring? Are you keeping those petri dishes at room temp?

Well...these plates are failures...VERY slow movement of mycelium and now trich has appeared.... oh well...glad all my eggs weren't in these baskets..

Although very slowly, the plate on the right side has made some progress. But yeah, I agree with you. Sometimes you just have to start over

Ok... I have a theory here..the antibiotic became concentrated in the bottom of my Erlenmeyer flask to the point of making my plates extremely hostile to bio-growth. Now...It's taken a couple weeks..but I have a set of genetics here that seems to be advancing...I think instead of throwing these plates away.. I might transfer to some fresh agar and check this sub-strain out because if it can survive such an environment, I would be interested to see the aggressiveness of colonization in grains, and also the fruits and potency will be awesome to test even if they are week.

Or you could just transfer it to a new plate like I suggested a week ago
